Object i ve:To discuss the expression and function of regulatory molecules B7-H3 and B7-H4 in laryngeal carcinoma,and to discuss the related mechanism.Method:1. The expression and distribution of the B7 family members and epithelial mesenchymal transition (EMT) markers in laryngeal carcinoma by immunohistochemistry.2. The co-expression the B7-H3 and EMT makers, B7-H4 and EMT markers in laryngeal carcinoma by double immunofluorescence method 3. Construction of B7-H3/B7-H4 plasmid and then transfected them into human epidermoid carcinoma cells(Hep-2); the expression of B7-H3/B7-H4 were detected by Western blot and q-PCR. Then compared the levels of B7-H3/B7-H4 between transfected and non-transfected Hep-2 cell.4. To test the invasion ability of transfected Hep-2 cell by transwell method and to test the transfer ability of transfected Hep-2 cell by wound-healing assay.Results:1.B7-H3 and B7-H4 were both expressed on the membrane and cytoplasm in human laryngeal carcinoma. In addition, B7-H3 was expressed on the tissue infiltrating capillary endothelial cells in human laryngeal carcinoma.B7-H4 was expressed on the tissue infiltrating lymphocytes in human laryngeal carcinoma.2. EMT markers were expressed in human laryngeal carcinoma.3. EMT markers and B7-H3 were co-expressed in human laryngeal carcinoma. EMT markers and B7-H4 were co-expressed in human laryngeal carcinoma.4. Compared with the non- transfected Hep-2, the expression levels of B7-H3 and B7-H4 in transfected Hep-2 were increased 5. Compared with the non-transfected Hep-2, the q-PCR results of B7-H3 and B7-H4 in transfected Hep-2 were increased. 6. The invasion ability and the transfer ability of transfected Hep-2 cell was significantly increased than non-transfection group.Conclusion:B7-H3 and B7-H4 were expressed in human laryngeal carcinoma. EMT markers and B7-H3 were co-expressed in human laryngeal carcinoma. EMT markers and B7-H4 were co-expressed in human laryngeal carcinoma. B7-H3 and B7-H4 were overexpressed in transfected Hep-2. The invasion ability of transfected Hep-2 cell was significantly increased. This indicated that the invasion and metastasis of laryngeal carcinoma were associated with the interaction between B7-H3/B7-H4 and EMT markers.
