The pathogen of Acquired Immunodeficiency Syndrome(AIDS)is Human immunodeficiency virus,which not only seriously threaten the people' s public health,but also influence the economic development and society stabilization.CEMĂ—174 cell line infected by SIV was used as a in vitro model for mechanism research of HIV interact with cells and anti-HIV research.The changes of immune regulation markers of cells post infection of SIV are not clear.In this research,CEMĂ—174 cell line was infected with SIVmac251 and the cell samples were collected dynamically.The mRNA of CD28 superfamily include CD28,ICOS,CTLA-4,BTLA,PD-1 and IDO,FOXP3 were tested with quantitative SYBR Green-based real-time PCR method. Additionally,SIVmRNA was also tested with the same method at the same time.The results showed that SIVmRNA was up-regulated significantly post 48h of SIV infection.Accompanied with the ascend of SIVmRNA,the mRNA of CD28,ICOS,CTLA-4,PD-1,IDO,FOXP3 also up-regulated in all but the last time point.In the last time point(216h post infection),the mRNA of all these immune regulation markers were not ascend or return.The mRNA of BTLA is the only immune regulation marker which don' t up-regulation corresponding to the SIVmRNA.The Chinese herb complex of supporting healthy Qi to eliminate pathogens was separated to herb complex of supporting healthy Qi and another herb complex of liminate pathogens.The effects of the three herb complexes to the mRNA of immune regulation markers in CEMĂ—174 cell line were researched.The research showed that the effects of all these three herb complexes to the SIVmRNA and immune regulation markers are superior to the AZT.It is notable that the Chinese herb never had been found to have the SIV inhibition effect superior to AZT.It is worthy of further research.Combined the above experiment results,the dynamic changes of mRNA of immune regulation markers in SIV infected CEMĂ—174 cell line were revealed originally.The SIV inhibition effect of Chinese herbs may not inferior to Chemical reagents.The research related to the SIV inhibition effect and mechanism,the active ingredients of the Chinese herb complex of supporting healthy Qi to eliminate pathogens must go on.
