Objective: To observe the distribution and expression of IL-21(Interleukin-21, IL-21) in laryngeal carcinoma tissue and adjacent normaltissue and explore the role of IL-21in the pathogenesis of laryngealcarcinoma. Methods: Clinical data of96cases of pathologically diagnosedlaryngeal carcinoma were collected in this study. RT-PCR and Western blotanalysis were used to detect the mRNA and protein expression of IL-21between the laryngeal carcinoma tissue and adjacent normal tissue.Immunohistochemistrical assay was employed to study the characteristic ofdistribution of IL-21in the laryngeal carcinoma tissue and adjacent normaltissue. Further, to evaluate the relationship of IL-21between the clinicalparameters and expression with laryngeal carcinoma. Results: RT-PCRshowed that the expression of IL-21was significantly increased inlaryngeal carcinoma, compared to adjacent normal tissues with statisticallysignificant (P<0.01).Western blot analysis further supported the similarresults in protein expression of IL-21in the laryngeal carcinoma tissues (P<0.01).The expression of IL-21closely related of the clinical parametersof tumor. Some cells showed very weak expression of IL-21in adjacent normal tissue. Compared with adjacent normal tissue, some tumor cells andinflammatory cells in laryngeal carcinoma tissue showed significantexpression of IL-21, and the strong expression and distribution of IL-21located in the nucleus and cytoplasm in the laryngeal carcinoma.Conclusion: The gene and protein expression of IL-21in laryngealcarcinoma were significantly increased closely associated with clinicalprogression, indicating that IL-21plays a role in the developed process oflaryngeal pathology; it may be as an important prognostic indicator inclinical judgment.
