The purpose:In this paper, puerarin(Pue)on inhibition of hypoxic pulmonary artey smooth muscle cells in rats (PASMCs) Pue-induced apoptosis and apoptosis molecular mechanism PASMCs,That Pue by regulating PI3K/AKT pathway plays the role.Methods:Train the first generation PASMCs,2-5can be administered on behalf of t he intervention and group training.Hypoxic cells were cultured in a conventional incubat or incubator. Experimental first observation of the effects Pue of hypoxia PASMCs apo ptosis.Serum starvation (SD) group was the control group,the cells were divided into fo ur groups:normoxic group(SD group),normoxia+Pue group,hypoxia group,hypoxia+Pue gr oup,culture24h.Vitro MTT(MTT)determine the effect of different concentrations under n ormoxic and hypoxic conditions in Pue on PASMCs vitality, cell morphology was obser ved by acridine orange staining,Using AnnexinV-FITC apoptosis detection kit to detect cell apoptosis,Determination of tumor tumor necrosis factor(TNF-α),Bax,Bcl-2and P-AK T expression by Westernblot law.Pue impact study PASMCs apoptosis signaling pathways.Experimental groups as follows: normoxic group(SD group).hypoxia group,hypoxia+Pue group,hypoxia+Pue+LY294002group,hypoxia+LY294002group.Pue administration concentration of250μmolL-1.AKT inhibitor (LY294002) concentration of10μmol·L-1,delivery time is24h.Westernblot assay using the PI3K/AKT pathway is involved in the inhibition of hypoxia-induced Pue PASMCs apoptosis process,Were determined expression of Bax,Bcl-2and P-AKT.Results:MTT assay showed, hypoxia can reduce apoptosis,inhibition of apoptosis P ASMCs. concentration of250μmol·L-1Pue effects beginning, Under normoxic conditio ns had no effect Pue on PASMCs apoptosis;Pue no effect on the expression of TNF-α under normoxic and hypoxic conditions.Pue can reverse hypoxia-induced Bcl-2(p<0.01) upregulation,Bax(p<0.01)downregulated.Under normoxic conditions Pue no protein kinase B (P-AKT)the expression of hypoxia so P-AKT(p<0.01)upregulation,Pue reversal of hy poxia-induced P-AKT(p<0.01)upregulated.LY294002and Pue can inhibit hypoxia-induced Bcl-2.Akt upregulated and downregulated Bax. Hypoxia or hypoxia alone dosing Pue a dded separately LY294002contrast,hypoxia+Pue+LY294002group Bcl-2,P-AKT more sig nificant downregulation.Bax upregulation more pronounced.Conclusion.Pue hypoxia inducible PASMCs inhibition of apoptosis,this effect may be through the PI3K/AKT pathway regulation.
