ObjectiveChoose extraction of puerarin in radix puerariae best extraction processconditions and purification process conditions; Investigation of extracts fromradix puerariae chain with urea bacteria element (STZ) of rats caused by thediabetes nephrosis kidney damage of protection.MethodsIn reference to the literature, and on the basis of the supercritical CO2fluidextraction in the extraction of puerarin puerarin,crushing the grain size forcertain functional materials, vacuum drying after extraction, concentrated crudeextractings get. In the influential factors in45℃temperature although is best, but in40℃temperature when theresults of itsvery close, for saving energy dissipation consideration, get the best extractiontechnology, extracting temperature is40℃,extracting pressure is35MPa,extracting time is4.5h, entrainment agents density is30%.Diabetic rats, blood sugar,24h urine protein increased significantly (P <0.01), and diabetes group, puerarin extract treatment group than rats bloodsugar,24h urine protein obvious reducing (P <0.05or P <0.01); Diabetic rats, renal hypertrophy index increased significantly, the puerarin extract kidneyhypertrophy treatment group extent lighter, statistically significant (P <0.05);Light first show that the rats diabetes group glomerular change, capillary thethickened basement membrane, luminal stenosis, glomerular staining area HEexpanded, pueraria after treatment of the above abnormal extract are differentdegree of improvement (P <0.05or0.01).ConclusionIn supercritical CO2fluid extraction (SFE–CO2) extraction of puerarinpuerarin, can finish at the same time distillation and extraction two process,extraction rate is high, the greatest degree keep effective ingredients, goodselectivity. By lowering blood sugar, pueraria lobata extract kidney hypertrophyindex, urine protein content of kidney pathological change and inhibition toimprove rats diabetes symptoms.
