Preventive Effect Of Jiedu Tongluo Fang On EAE Rats And The Study On The Effects Of MMP-9

Objective: By constructuring acute experimental autoimmune encephalomyelitis of rats model, Observeing Bu Shen Tong Luo Formula can effectively prevent the incidence of EAE, and research part of the mechnism of Jie Du Tong Luo Formula for preventive treating acute EAE rats, which provide experimental basis for clinical practice.Methods: Using Guinea pig spinal cord homogenate two-induced immune Wistar rats, to contruct acute EAE model, By using different dos of Bu Shen Tong Luo Formula for preventive treating, observing the morbidity, recording clinical score and weight changes, and killing the rats in acute and convalescence stage. Through H.E. staining and myelin staining, observing patho change; level of MMP-9 in CNS was measured by streptavidin perosidase.Results:The mobility and neurological function of the rats: in the 7th day, no rats has the attack in the control group.The differences, in the mobility and neurology between model group, hormone group and the Chinese medicine group, have no difference.Each rat in the EAE group lost weight,and the relative ratios have no difference14days later, after building the EAE model,the neurological score of the rats: compared with the hormone group,the Chinese medicine group decreased slightly. Compared with the model group, the Chinese medicine group and the hormone group both decreased obviously. The amount of the inflammatory cells in histopathological slices: Chinese medicine group is slightly lower than the hormone group. Compared with the model group, the Chinese medicine group and the hormone group both decreased obviously. The expression of the MMP-9: the difference between the Chinese medicine group, hormone group and the model group are both significant. There is nearly no difference between the Chinese medicine group and the hormone group.21 days later,he neurological score of the rats: compared with the hormone group,the Chinese medicine group decreased significantly. Compared with the model group, the Chinese medicine group and the hormone group both decreased obviously. The difference between the Chinese medicine group and the hormone group are significant.Conclusion:(1)The EAE Wistar rat model induced by guinea pig spinal cord homogenate which added complete freund’s adjuvant,joint with inactivated attenuated bordetella pertussis,is stable, reliable, and has high incidence.(2) 14 days later, which is at the acute phase, the expression of the MMP-9 in the model group is higher than the control group. And both the hormone group and the Chinese medicine group are obviously lower than the model group. 21 days, the catabatic period, compared with the hormone group, the Chinese medicine group is lower. All the differences above have statistical significance.The demoving toxin and activating collaterals decoction can decrease the inflammatory reaction in the central nervous system of the EAE rats, significantly. The mechanism of inducing the nerve injury of the EAE rats may be by inhibiting MMP-9 expression and decreasing the damage to the blood brain barrier in the acute stage, thus protect the nerve function of the rat.(4) The demoving toxin and activating collaterals decoction is used in the secure therapy of multiple sclerosis in the acute phase, has the long-term curative effect.
