Objective: application of APP transgenic mouse model of dementia as experimental animal model, the traditional Chinese medicine Huoxue Kaiqiao prescription on the APP dementia mouse model of intervention, pathology analysis before and after the intervention behavior, the study of traditional Chinese medicine Huoxue Kaiqiao effects of prescription in brains of APP transgenic mice into dementia model of A beta pathway and cholinergic system, serum inflammatory factors and other related indicators, to explore traditional Chinese medicine Huoxue Kaiqiao prescription and the mechanism of action of AD transgenic mice model, and provide the basis for the deepening of clinical application.Methods: the March old APP transgenic mice, half male and half female, were randomly divided into large dose of Chinese medicine group and Chinese medicine small dose group and donepezil hydrochloride group, model group, use the same genetic background of non transgenic mice(C57BL/6J mice) as a normal control group, with 10 rats in each group, continuous administration changes after 3 months Morris water maze experiment of APP transgenic mice learning and memory ability, immunohistochemistry in hippocampus of mice within Ch AT, Ach E, A beta 1-42, beta-APP, caspase-3 expression, enzyme linked immunosorbent assay in mice serum IL-1, IL-6, TNF- alpha content. Discussion on traditional Chinese medicine Huoxue Kaiqiao prescription and the mechanism of action of AD transgenic mice model.Results: the Morris water maze test results show that: compared with the normal control group, model group mice in June age through the original number of platform location, fourth quadrant swimming time and distance shortened significantly and reduce(P<0.01 orP<0.05); after treatment, mice in the Morris water through many officials quadrant swimming time and distance of fourth original position of platform the number of times, compared with the model group increased significantly(P<0.05 or P<0.01). Compared with normal control group, model number of Ch AT positive cells in hippocampal CA1 area of mice and area ratio was significantly decreased(P<0.01), the number of Ach E positive cells and the area proportion increased significantly(P<0.01), compared with the model group, the number of Ch AT positive cells to the drug group and area ratio improved significantly(P<0.05 or P<0.01), Ach E positive cell number and area ratio was significantly decreased(P<0.05 or P<0.01). Compared with the normal control group, model group mice hippocampus CA1 area A beta 1-42 and beta-APP, the number of Caspase-3 positive cells and the area proportion increased significantly(P<0.01), compared with model group, each group after treatment, the positive cell numbers and area ratio was significantly decreased(P<0.05 or P<0.01). Compared with the normal control group, the serum IL-1 and IL-6 in model mice, TNFcontent was significantly increased(P<0.01), Compared with model group, each group after treatment the serum levels of IL-1, IL-6, TNF- content was significantly decreased(P<0.05 or P<0.01).Conclusion: Traditional Chinese medicine Huoxue Kaiqiao component can be improved to some extent in APP transgenic mice, improve the ability of learning and memory of mice hippocampus Ch AT expression, decreased Ach E expression, decreased in mice hippocampus A beta 1-42, beta-APP, caspase-3 expression, reduce serum inflammatory factor levels.
