| Gastric cancer means the malignant tumor of occurrence in gastric mucosal epithelial cells.Our country is the high incidence area of gastric cancer,the morbidity and mortality were living in a variety of malignant tumors of the anterior.RecentlyTreatment in resection of gastric cancer and chemical drugs despite great progress, but does not make long-term survival rate improved, recurrence and metastasis of tumor is the ultimate cause of deathin patients.A large number of clinical research data show that, gastric cancer cured after 2 years, the recurrence and metastasis are almost straight up,the total relapse rate is more than 70% among two years.Therefore, the most important factor to prolong the survival of patients is to reduce the recurrence and metastasis of gastric cancer rate and also the difficult problems of medical research field.Part 1 Therapeutic Effect of Weiliuan on gastric tumor in nude mice and the effect of CXCL4 and CXCL12We do the research by the following appraoches to Explore Effect of Weiliuan on gastric tumor in H22 sarcoma mice immune system.,(1)T lymphocyte esterase staining rate.(2) delayed type hypersensitivity(3)hemolysin antibody(4)Phagocytic Clearance Ability.The results showed that (1) the number of T lymphocytes increased significantly after the demolition Weiliuan and splited drugs, H22 solid tumor in mice decreased significantly than the control group; (2) mice ear delayed hypersensitivity intensity of Weiliuan and splited Ⅰ and Ⅱ significantly enhanced; (3)Weiliuan and splited Ⅰ and Ⅱ group could obviously promote the formation of hemolysin antibody. (4) The phagocyticcoefficient demolition of Splited II group increased significantly, while the other groups showed a tendency to increase, but no obvious change.Part 3 effect of Weiliuan on gastrointestinal function restorationThe rats were given the decoction of Radix et Rhizoma combine with dietary disorder to induce spleen-deficiency syndrome.after treatment of Weiliuan,Dopamine was injected into rats’ abdominal cavity;The charcoal propulsion rate and the gastric residual rate were calculated;The influence of different formulas of Weiliuan on Dopamine in restraining peristole of rats of spleen deficiency;Rats’abdominal cavity was injected into proserine;And the influence of Weiliuan on intestine propulsive role.Weiliuan group,Weiliuan splited prescription I group and Weiliuan splited prescription II group can significantly reduce the spleen index,the level of SDH and LDH,it can also improve the level of the AMSIn the regulation of hormone levels, Weiliuan group,Weiliuan splited prescription I group and Weiliuan splited prescription II group can significantly improve the levels MTL、GAS,but had no obvious effect on SS.Part 4 The effect of Weiliuan on Blood rheology of H22 ratsWe also explore the effect of Weiliuan on Blood rheology of H22 rats stasis rats can Significantly reduce the value of whole blood viscosity and erythrocyte aggregation index caused by Adrenaline and cold;splited II can Significantly reduce the value of whole blood viscosity and erythrocyte aggregation index when In the shear rate was 30,5,1Part 5 Effect of Weiliuan on cell proliferation and cell cycle in gastric cancer cells Subculture was carried on by Human Gastric Carcinoma Cell Line SGC-7901 in RPMI 1640 culture solution. MTT assay was used to determine the inhibitory effect of Weiliuan on the growth of SGC-7901 cells. The changes of cell cycle of SGC-7901 were detected by flow cytometry. MTT assay show that Weiliuan could inhibit the proliferation of cell line SGC-7901,cell cycle analysis indicated that Weiliuan can arrest SGC-7901 at S phase,both of them were time-and dose-dependent manner.:Weiliuan can inhibit proliferation of gastric carcinoma cells,and it can arrest cell cycle of S phase may be one of the mechanisms. |
