Objective:By determining the MK-801 induced schizophrenia model rats’serum SOD activity and the content of MDA and NO,explore the effect Shiweiwendantang on oxidative stress injury and provide experimental basis and support for clinical medicine,and explore the significance of treatment on schizophrenia with Chinese Medicine"insufficiency of phlegm method"Methods:40 clean SD rats were randomly divided into blank group (group A),model group(group B),control group (group C) and the treatment group(group D),10 in each group.First stage:model preparation. Group A:intraperitioneal injection of saline;group B,C and D:intraperitioneal injection of MK-801(0.1mg/kg),inject 14 days continuously.Second stage:therapeutic medication.On the 15 days of preparing model,group A and B were dosed with distilled water,group C were dosed with Shiweiwendantang(12.15g/kg),group D were dosed with suspension risperidone (0.036mg/kg),dose 14 days continuously.Third stage:indicators determination.On the 15 days of dosing,sacrificed the rats by anesthesia,open the chest,collect blood through cardioid.After the treatment,using methods xanthine oxidase (WST-1),thiobarbituric acid method (TBA),solid phase sandwich enzyme-linked immunosorbent assay (ELISA) respectively to determine the experimental rats’ serum SOD activity and the content of MDA and NO.Results:1.In this experiment,model group rats’ serum SOD activity was significantly lower than blank group(P<0.01),the content of MDA was higher than the blank group(P<0.01),it signed that inner of MK-801 induced model rats exist the oxidative stress injury;in the control group and the treatment group rats’serum SOD activity was significantly higher than the model group(P<0.01),the content of MDA was lower than the model group(P<0.01),indicating Shiweiwendantang and risperidone can increase model rats’content of anti-oxidants, remove excess oxygen free radicals,have the effect on resisting oxidative stress.2.The content of NO in rats’ serum of the model group were significantly higher than the control group(P<0.01),indicated that there’s abnormal NO-NOS system in the MK-801 induced schizophrenia rats;in control group and the treatment group the content of NO in rats’serum was lower than the model group(P<0.01),described Shiweiwendantang and risperidone may prevent NO glutamate-mediated neurotoxicity system by reducing the content of NO in model rats’serum to achieve therapeutic effect.3.There’s no statistical differences on Rats’serum SOD activity,the content of MDA and NO between the control group and the treatment group(P>0.05),described Shiweiwendantang and risperidone had the effect on the mechanism of regulating the oxidative stress of the schizophrenia animal model.Conclusion:Shiweiwendantang can significantly increase the MK-801 induced schizophrenia model rats’serum SOD activity while reduce the content of the MDA and NO.The mechanism of this formula may correlative with increasing model rats’content of anti-oxidants,removing excess oxygen free radicals,reducing lipid peroxidation damage,increasing the ability of resisting oxidative stress and regulating the abnormal NO-NOS system,stop NO mediated neurotoxic effects.
