Obj ective:In this experiment,schizophrenia maleate(MK-801)was used to establish schizophrenia model rats.After modeling and treatm ent,behavioral experiments were performed.Then,serum CAT activ ity,SOD activity,GSH-Px activity,as well as MDA and NO conte nt,explored the role of Fuyuankang Capsule in regulating oxidati ve stress injury in schizophrenia model rats.Methods:Forty male SD rats of clean grade were divided into blank group,model group,control group and Chinese medicine group according to random grouping method,with 10 rats in each group.The blank group was intraperitoneally injected with normal saline.The model group,the control group and the Chinese medicine group were intraperitoneally injected with MK-801 at a dose of O.lmg/kg.After 14 days of continuous inj ection,behavioral observation was performed to verify the establishment of an animal model of schizophrenia.On the 15 th day of the model,the therapeutic administration was carried out.The blank group and the model group were given distilled water;the control group was administered with the risperidone suspension at a dose of 0.03 6mg/kg;the Chinese medicine group was administered with the dosage of 0.58g/kg.The capsule suspension was continuously administered for 14 days and then subjected to behavioral observation.On the 15th day,blood was taken from the rat heart.After treatment,the CAT activity,SOD activity,GSH-Px activity,MDA content and NO content in the serum of the rats were tested by ELISA.Results:1.MK-801 induced model rats in the open field experiment,the total distance of movement decreased,the number of groomi ng increased,indicating that the exploratory activity of rats decr eased,anxiety behavior increased;in the Morris water maze exper iment,the escape latency extended,through the original platformThe decrease in the number of times indicates that the learning and memory and spatial localization memory ability of the mod el rats are reduced.After the drug treatment in the control grou p and the Chinese medicine group,the behavioral symptoms of t he above-mentioned test rats are improved.2.The CAT activity,SOD activity and GSH-Px activity in the serum of the experimental model group were significantly lowe r than those in the blank group(P<0.01),and the MD A content and NO content were significantly higher than the blank group(P<0.01),suggesting that MK-801 Oxidative stress damage is pres ent in the model rats of schizophrenia.3.Compared with the model group,serum CAT activity,SOD activity and GSH-Px activity in the control group and the Chinese medicine group were significantly increased(P<0.01),MDA content and NO content were significantly decreased(P<0.05,P<0.01).It is indicated that Fuyuankang Capsule and Risperidone can exert antioxidant effects by regulating oxidative stress damage.4.Compared with the traditional Chinese medicine group,there was no significant difference in serum CAT activity,SOD activity and MDA content between the two groups(P>0.05).The GSH-Px activity in the Chinese medicine group was higher than that in the control group(P>0.O5).Lower than the control group(P<0.01),suggesting that Fuyuankang Capsule and risperidone have similar effects in regulating oxidative stress injury in schizophrenia model rats.Conclusion:Fuyuankang Capsule can significantly increase CAT activity,SOD activity,GSH-Px activity and decrease MDA content and N O content in schizophrenia model rats.It shows that Fuyuankang Capsule has the function of improving antioxidant capacity and scavenging excess free radicals.
