ObjectiveBy researching the expression of hMSH2 and Ki-67 in ovarian epithelial cancer, ovarian epithelial benign tumor and normal ovarian epithelial tissues, and the respectively expression of hMSH2, Ki-67 in epithelial ovarian cancer, different histological types, different surgical staging and different pathological grades, and also study the relevance with digenic, explore the functions of hMSH2 and Ki-67 on the occurrence and development of epithelial ovarian cancer to provide a theoretical reference for epithelial ovarian cancer diagnosis and prognosis.Method84 epithelial tissue specimens of ovary (paraffin block archived by Pathology Department, and complete clinical data), removed by surgery, of gynecological inpatients admitted in the First Affiliated Hospital of Kunming Medical University from November 2011 to May 2014 were collected, including 28 cases of normal ovarian tissue,28 cases of epithelial benign tumor of ovary and 28 cases of epithelial cancer of ovary.Patients with epithelial benign tumor of ovary included 14 cases of serous cystadenoma and 14 cases of mucinous cystadenoma. Patients with epithelial cancer of ovary included 12 cases of serous cystadenocarcinoma,8 cases of mucinous cystadenocarcinoma and 8 cases of endometrioid adenocarcinoma. Patients with epithelial cancer of ovary included 12 cases in phase I-II and 16 cases in phase III-IV based on surgery and pathology staging according to 2000 International Federation of Gynecology and Obstetrics (FIGO) standard. Patients with epithelial cancer of ovary included 20 cases of grade G1-G2 and 8 cases of grade G3 based on grading standards of WHO grading. All specimens were fixed with 10% formalin and embedded by regular paraffin and made as 4um thick slices. They were subject to HE staining and definite pathological diagnosis.Use immunohistochemical Envision method to detect the expression of hMSH2 and Ki-67 in 28 cases of normal ovarian tissue,28 cases of benign ovarian tumors and 28 cases of epithelial ovarian cancer tissues, and apply SPSS 17.0 software package for statistical analysis, use group to design diversity sample comparative rank and use Kruskal-Wallis H test method, two-sample comparison rank, Mann-Whitney U test method, and Spearman rank correlation analysis for statistical academic treatment.Results1. The positive expression rate of hMSH2 in normal ovarian epithelial tissue, epithelial benign ovarian and epithelial ovarian cancer was:46%,56%,96%; the positive expression of hMSH2 in epithelial ovarian cancer group was significantly higher than in normal ovarian tissue and epithelial benign ovarian tissues (P<0.05), whereas the expression level of hMSH2 in normal ovarian tissue and epithelial benign ovarian had no significant difference; the positive expression level of hMSH2 in epithelial ovarian cancer raised with the increase of surgery and pathology staging (P<0.05), and raised with the reduction of histological differentiated degree (P <0.05), but it was regardless of histological type.2. The positive expression rate of Ki-67 in normal epithelial ovarian tissue, epithelial benign ovarian expression and epithelial ovarian cancer was:0%,7%,93%; the positive expression of Ki-67 in epithelial ovarian cancer group was significantly higher than in normal ovarian tissue and epithelial benign ovarian tissues (P<0.05), whereas the expression level differences of Ki-67 in normal ovarian tissue and epithelial benign ovarian had no statistical significance; the positive expression level of Ki-67 raised with the increase of surgery pathology staging (P<0.05), and raised with the reduction of histological differentiated degree (P<0.05), but it was regardless of histological type.3. The expression of hMSH2 and Ki-67 was positively correlated in epithelial ovarian cancer, r= 0.469, P= 0.012 (<0.05);Conclusions1. The expression level ofhMSH2in epithelial ovarian cancer is higher than normal ovarian tissue and epithelial ovarian benign which explained that hMSH2 plays an important role in the occurrence of epithelial ovarian cancer, the positive expression level of hMSH2 in epithelial ovarian cancer had nothing to do with histological type, but it related with surgery and pathology staging and pathological grading; with the increase of surgery and pathology staging and the reduction of histological differentiation degree, the positive expression of hMSH2 increased, indicating thathMSH2 plays an important role on the development and metastatic malignant biological behavior of epithelial ovarian cancer.2. The positive expression level of 2. Ki-67 in epithelial ovarian cancer is higher than normal ovarian tissue and epithelial ovarian benign which explained that Ki-67 plays an important role in the occurrence of epithelial ovarian cancer, the positive expression level of Ki-67 in epithelial ovarian cancer had nothing to do with histological type, but it related with surgery and pathology staging and pathological grading; with the increase of surgery and pathology staging and the reduction of histological differentiation degree, the positive expression of Ki-67 increased, indicating that with the increase of clinical stage and the reduction of pathological tissue differentiation degree in epithelial ovarian cancer, the evolution of the entire malignant biological behavior, the proliferation of malignant cells become more active, which result to more prone to invasion and metastasis.3. The expression of hMSH2 and Ki-67 has a positive correlation which indicates that with the proliferation of cancer cells, the rate of DNA mismatches risk increases, and the expression compensatory of hMSH2 rise, these two things play a synergistic role in the occurrence, development and prognosis of epithelial ovarian cancer; for the patient with high expression of hMSH2 and Ki-67 may indicate a high degree of malignancy.
