Backgrounds and objectivesOvarian cancer (ovarian cancer, OC) is one of the familiar vicioust tumor ofgynaecology. The top of the case fatality rate in in gynecologic malignant tumor isovarian cancer.After the incidence of cervical cancer and uterine cancer, Ovariancancer is the third largest in gynecological malignancies. Because of patients withearly symptoms and signs are not obvious, the short of early diagnosis methods,resulting in about70%patients diagnosed in the advanced stage. Only25%ofpatients diagnosed in stage I, survival rate of stage III and IV ovarian cancer patientswas20%to25%, however the survival rate of stage I was95%. At present thedevelopment of ovarian cancer mechanism is unknown so far. Insulin-like growthfactor mRNA-binding proteins(IMPs) have three similar proteins, which are namelyInsulin-like growth factor mRNA-binding protein1(IMP1), IMP2, IMP3. IMPs cancombine and code mRNA of IGF II highly specificly,which participate in adjustingthe location, stability and reverse transcription and translation of IGF II mRNA.Theimbalance of IGFII mRNA stability and function was occurred in thepost-transcriptional level,which can give rise to IGFII overexpression that can prompt cancerization.The research proved that the expression of IGFIImRNA in epithelialovarian cancer was high.But the researches of the expression of IMP1,as one of IGFIItranscription regulation factors,and the relationship between IMP1and the occurrenceand development of ovarian cancer were seldom.This study is aimed of discussing theexpression condition of IMP1in epithelial ovarian cancer and furtherunderstanding the mechanism of genesis and developmen of epithelial ovariancancer.Material and Methods:1Clinical dataNinty five cases were chosen from September2011to march2013from the firstaffiliated hospital and the third affiliated hospital of Zhengzhou University.The nintyfive cases confirmed by pathological science were forty seven cases with epithelialovarian cancer and twenty eight cases with epithelial ovarian benign tumor andtwenty cases with normal ovarian epithelial tissue. Normal ovarian tissues were frompatients of the uterus disease and breast disease with hysterectomy andhysterectomy and bilateral adnexectomy.The forty seven cases with epithelial ovariancancer were thirty two serous cystadenocarcinoma and ten cases with mucouscystadenocarcinoma and five cases with endometrial carcinoma.The ages of theepithelial ovarian patients were twenty nine~seventy six,the median age was fiftyone.According to the international union of gynecology and obstetrics staging, thecases of epithelial ovarian cancer were covered eighteen cases of I~II period andtwenty nine cases of III~IVperiod, twenty seven cases with high differentiationand twenty cases with poor differentiation; nineteen cases with lymph nodemetastasis and twenty eghit cases without lymph node metastasis;twenty seven caseswith ascites and twenty cases without ascites;All cases preoperative were withoutradiotherapy and chemotherapy. All specimens after in vitro were quickly cut into twocopies,one copy after liquid nitrogen frozen was saved into-80℃refrigerator,theother one after formaldehyde fixed by10%neutral and paraffin embedding was cutinto3um,which was used for the immunohistochemistry. 2Experimental MethodsImmunohistochemistry of biotin Streptomyces protein peroxidase (SP) methodwas used to measure the protein expression level of IMP1in47cases of epithelialovarian cancer and28cases of epithelial ovarian benign tumor and20cases ofnormal ovarian epithelial tissue.Relations between the protein expression levels ofIMP1in the epithelial ovarian cancer and the clinical-pathological prognosticcharacterisitics of epithelial ovarian cancer were analyzed statistically. The RT-PCRtechnology was adopted to detect IMP1mRNA expression of47cases of epithelialovarian cancer and28cases of epithelial ovarian benign tumor and20cases ofnormal ovarian epithelial tissue. Relations between the IMP1mRNA expressionlevels and the IMP1protein expression levels in the epithelial ovarian cancer wereanalyzed statistically.3Statistical analysisAll dates were analysed by SPSS17.0statistical software, and the results in theform ofx s. Qualitative data were analyzed by χ2test, and quantitative data byone-way ANOVA (homogeneity of variance) or the Kruskal-Wallis test (for unequalvariances), Bonferron methods were used in comparison between groups. Spearmanrank correlation was used in correlation analysis. Statistical significance level was0.05.Results:1.The expression of IMP1protein in epithelial ovarian cancerThe positive expression rate of IMP1protein in the epithelial ovarian cancer orepithelial ovarian benign tumor or normal ovarian epithelial tissue was5.1%(40/47),32.1%(9/28),10%(2/20) respectively.The IMP1protein expression in the epithelialovarian was significantly higher than epithelial ovarian benign tumor and normalovarian epithelial tissue(χ2=39.239,P=0.000).,it was no significant differencebetween the epithelial ovarian benign tumor and normal ovarian epithelialtissues(P=0.147). 2.the relationship between IMP1protein and the clinical and pathologicalfeatures of epithelial ovarian cancerThe expression of IMP1protein in epithelial ovarian cancer was associated withFIGO stage and histological grade, It is a significantly difference between III~IVovarian cancer and I~II ovarian cancer (χ2=5.645,P=0.018).The positive expressionrate of IMP1protein in poorly differentiated epithelial ovarian carcinoma wassignificantly higher than in moderately and high differentiated epithelial ovariancarcinoma(χ2=4.365,P=0.037).The expression had nothing to do with lymph nodemetastasis and presence of ascites (P>0.05).3.RT-PCR resultThe478bp amplification bands of β-actin and334bp amplification bandsof IMP1were visible in95cases of ovarian cancer.The relative expression values ofIMP1in the normol ovarian tissue, epithelial ovarian benign tumor tissue, epithelialovarian cancer tissues were0.085±0.011,0.120±0.013,0.652±0.067respectively,thenwere enhanced in turn.The values of ovarian cancer groupthere were significantlydifference beteen normol ovarian group and epithelial ovarian benign tumor group((F=990.674, P=990.674),there was no statistically significant difference expression (P=0.058).4. Correction analysis of IMP1protein and IMP1mRNAUsing Spearman rank correlation,it is analyzed the correction of IMP1proteinand IMP1mRNA in47cases of epithelial ovarian cancer. It was found that the resultsof immunohistochemical staining and RT-PCR (mRNA levels) were positivelycorrelated (correlation coefficient r=0.869, P=0.869),that it was highly correlatedbetween the IMP1protein and mRNA.Conclusions:1. IMP1protein and mRNA expression was abnormaly high in epithelial ovariancancer and associated with FIGO clinical stage and histological grade of epithelial ovarian cancer, it may play an important role in the development and prognosis ofepithelial ovarian cancer.2. IMP1may be a potential diagnosis and therapeutic target of the epithelialovarian cancer.
