Dangshen, campanulaceae plants, is the dry root of Codonopisis pilosula (Franch.) Nannf, Codonopsis pilosula Nannf. Var. Modesta (Nannf.) L.T. shen or Codonopisis tangshen Olive. It has many effects, such as strengthing spleen and tonifying lung, nourishing fluid. If someone has the symptoms, include insufficiency of Qi, eat less fatigue, cough, asthma, lack of blood, pale complexion, palpitations, shortness of breath, fluid and’thirst, heat diabetes, he or she can eat Dangshen for treatment.This topic evaluates the traditional quality of GanSu Dangshen, and studies the preparation technology, quality standard, stability, vitro dissolution and the vivo pharmacokinetics of superfine powder of codonopsis pilosula. The result show superfine powder of codonopsis pilosula, the bioavailability will greatly increase, thereby save medicine resources.Literature studies the recent research progress of codonopsis, summarizes the resources distribution, characters, processing, commodity markets of Dangshen, verifies its history of use, analyzes and sum up its main chemical components, pharmacological actions, clinical applications, quality control situation, cultivation and storage of plant conservation status. And it also introduces superfine grinding technology principles, characteristics and application.After evaluating the traditional quality of GanSu Dangshen, it turned out that the content of the Lobetyolin and polysaccharides of Gansu codonopsis produced in Gansu Wen country is much higher than others, and also the extract content is higher. The preparation of superfine powder of codonopsis pilosula, optimization by orthogonal experiment and the preparation technology is determined:Dangshen about lkg, with ordinary mill crushed into meal spare (about the 4th through sieve), and then put these meals ultrafine mill, continuous grinding 40 minutes. According to the above identified route, we can produce three batches of superfine powder of codonopsis pilosula. Complete the production of ultrafine particle size measurement, the average particle size of 7.133μm, and determination of the wall-broken Rate, is 90.74%.The study set the quality standard of superfine powder of codonopsis pilosula, including character, identification, inspection, measuring and fingerprint. Established HPLC method for the determination study of lobetyolin:Thermo Hypersil Gold-C18 column (150×4.6mm,5μm); acetonitrile-water (20:80) as the mobile phase, detection wavelength 268nm.And to develop the product lobetyolin content of not less than 0.16%.Using UV spectrophotometry content dangshen polysaccharides:detection wavelength 490nm. And to develop the product polysaccharide contents is not less than 10.0%.The study also established the methods about the standard control fingerprint and fingerprint detection of superfine powder of codonopsis pilosula:octadecylsilane bonded silica as a filler (Column:Thermo Hypersil Gold-C18:150×4.6mm,5μm); flow rate of 1mL·min-1; detection wavelength of 268nm; acetonitrile as the mobile phase A, water as the mobile phase B, gradient elution.The study of stability of superfine powder of codonopsis pilosula shows that the stability of ultrafine Dangshen to superfine quality standards as a basis for three batches of ultrafine do long-term and accelerated stability test done research, completed six months accelerated testing, long-term stability studies are carried out to the first months. After accelerating tank stay like 6 months,10 months of retained samples at room temperature stability of the test, compared with October 2007, and no significant changes in the product description within 10 months of consistent quality.The vitro dissolution study shows that ultrafine Dangshen dissolution rate Lobetyolin and polysaccharides is far greater than dangshen ordinary powder. By comparing bioavailability of superfine powder in vivo of rats, the results showed that the bioavailability of its Dangshen ultrafine powder of 1.40 times, increased bioavailability of 39.7%. It can achieve the purpose of saving medicines. The Codonopsis fingerprint detection of superfine powder of codonopsis pilosula and general ultrafine powder showed that the characteristic peaks in the chromatogram does not increase nor decrease, but the peak area changes. This means that ultrafine grinding to a powder particle size is reduced, the original components of the dissolution rate and quantity are increased, and it did not change the original medicinal ingredients. Therefore, it could not produce toxic side effects in principle.
