Objective Analysis of Pangasisus haniltoa skin nutrition, extracted collagen from the fish skin and its physical properties, chemical properties analysis, and physiological characteristics were studied. Methods Respectively by atmospheric pressure drying hair moisture determination, according to Chinese pharmacopoeia2010 edition of annex VIII N determination of ash content, sox let extraction method to determinate crude fat content, semi-micro kjeldahl nitrogen determination method for determination of protein content. Acid-soluble collagen(ASC) and pepsin-soluble collagen(PSC) from fresh Pangasisus haniltoa skin; Using SDS-page gel electrophoresis and HPLC analysis to compare different molecular weight and amino acid composition of collagen;Use viscometry to determine Pangasisus haniltoa skin denaturation temperature and compare to other sources of collagen; For a variety of sources of collagen differential thermal analysis and comparison between different collagen thermal stability. Collagen sponge were separately measured water imbibition,dry weight loss rate and water solubility observe its physical properties; Determination of acidity and heavy metals content determination to observe it as a hemostatic sponge feasibility, through rabbit ear bleeding and liver bleeding experiment to explore the Pangasisus haniltoa skin references as a hemostatic sponge material. Results Pangasisus haniltoa skin moisture content was 66.14%, the ash content is 0.76%,Pangasisus haniltoa skin raw materials, through a treatment and after two the crude fat content were determined respectively, the results were 18.20%, 9.6%, 7.46%, crude protein content determination results were 87.29%, 90.73% and 95.20% respectively.Pangasisus haniltoa skin with ASC extraction yield is 58.2%, with pig mucosa pepsin enzyme hydrolysis the PSC extraction yield is 22.0%. The collagen protein electrophoresis determination results and collagen amino acid determination results show that the several different sources of collagen mainly for type I collagen,and the Pangasisus haniltoa skin on have the highest hydroxyproline and proline content in several kinds of materials, means that the most abundant collagen content; Pangasisus haniltoa skin viscosity measurement results show that the skin without any cross-linked by EDC crosslinking denaturation temperature is 34 ℃ or so, and after 120 ℃ hot crosslinking of Pangasisus haniltoa skin denaturation temperature instead of only28 ℃ or so. By differential thermal analysis showed that it also has good heatresistance. The study on the sponge made by Pangasisus haniltoa skin show that the performance of its physical properties and chemical properties are development become the characteristics of the hemostatic materials, the hemostatic experiments on animals more verify the Pangasisus haniltoa skin references on the sponge has excellent hemostatic effect. Conclusion The protein content on Pangasisus haniltoa skin is very rich, the preparation of hemostatic sponge has excellent hemostatic effect,can be further development into medical hemostatic materials used for clinical purposes.
