BackgroundUncontrollable bleeding(UB)is an unavoidable scenario in various body safety accidents and surgical operations.Consequently,how to control bleeding is also a focus area that the medical community has been paying attention to and continuously improving.At present,hemostatic materials have developed new forms of hemostasis such as light-crosslinking hydrogels to seal wounds.However,so far,the most widely used and mature hemostatic materials in the medical field are gauze,gelatin hemostatic sponge,chitosan hemostatic powder,external freeze-dried human fibrin adhesive,regenerated oxidized cellulose and other hemostatic agent through natural extracts manufactured products.Among them,hemostatic sponge is the second most used medical hemostatic material after gauze.An ideal hemostatic sponge should have the characteristics of natural biological sources,cheap raw materials,excellent hemostatic effect,good biocompatibility,and superior degradability.Cellulose is considered to be the most abundant organic polysaccharide polymer in the nature,and chitin is considered to be the second most abundant natural organic polysaccharide polymer.The application of sponges made of chitin,cellulose and their derivatives in the field of hemostasis has gained multiple studies,but the application of composite sponges with chitin and cellulose-related extracts in the field of hemostasis has not yet been studied.Therefore,the purpose of this study was to explore how to use chitin and corn stalk pith extract to fabricate a composite hemostatic sponge,and optimize it to obtain suitable physical characterization,as well as obtain the best calcium ion absorption capacity and hemostatic performance.Finally,we hope the study would provide a theoretical basis for the clinical application of the chitin-corn stalk pith composite sponge for rapid hemostasis.Objectives1.To study the synthesis method of chitin-corn stalk pith extract composite sponge,the change of physical characterization and biocompatibility of sponge2.To explore the calcium ion absorption mechanism of chitin-corn stalk pith extract composite hemostatic sponge and its procoagulant mechanism.Methods1.Preparation of composite hemostatic sponge:Chitin and corn stalk pith extract were dissolved in NaOH/urea solution by repeated freeze-thaw swelling,and then chitin was mixed with glutaraldehyde(GA)as a cross-linking agent.The hydrogel is obtained by cross-linking with the corn stalk Shuxin extract,and then freeze-dried to obtain a composite hemostatic sponge.2.Evaluation of composite hemostatic sponge characterization and performance indicators:scanning electron microscopy(SEM),swelling,mechanical properties,blood coagulation index(BCI),in vitro degradation experiments,mouse liver hemostasis experiments,etc.Results1.Chitin and corn stalk pith extract can be cross-linked by glutaraldehyde in NaOH/urea system.2.When the mass ratio of chitin-corn stalk pith core composite hemostatic sponge is 1:1,the best porosity,the largest wetting rate,the smallest elastic modulus and the best degradation rate are obtained.3.The introduction of corn stalk pith significantly increased the calcium ion absorption capacity of the composite hemostatic sponge,which promotes the activation of platelets and achieves a more superior hemostatic performance.Conclusions1.When the mass ratio of chitin and corn stalk pith extract is 1:1,the composite hemostatic sponge has the best porosity ratio,the largest swelling ratio,the smallest elastic modulus,and the best degradation characteristics for hemostatic application.2.The introduction of the corn stalk pith extract improves the absorption capacity of the hemostatic sponge to calcium ions,and subsequently obtains better platelet activation effect and hemostatic effect.Compared with the polyvinyl formaldehyde sponge,the chitin-corn stalk pith composite sponge owns better hemostatic properties,biocompatibility,and biodegradability than PVF sponge.
