Sub-health has become one of the major harm to human health. It is a serious impact onhuman life. Therefore, to find a scientific and feasible way to improve the symptoms ofsub-health become a research focus in the field. Chinese medicine has unique advantage in thetreatment of sub-health. In this study, the effects of Lycium barbarum polysaccharide on sleeping,antioxidant, immuno-enhancement and anti-fatigue were explored in sub-health model mice builtusing force-swimming, sleep deprivation, bounding and environmental stress. In theexperimental process, we defined and evaluated sub-health mice model according to the behavior,fatigue degree, biochemical index and pathological tissue sections. After the intervention ofLycium barbarum polysaccharide for21d, the blood routine, blood biochemistry, the activitiesof SOD, CAT and MDA level were detected. Sleep duration was calculated by intraperitonealinjection of pentobarbital sodium with threshold dose. The differences in thymus index andspleen index were evaluated by weighting the thymus and spleen. CD4+/CD8+ratio was detectedby flow cytometry technique and the content of5-HT and DA in hypothalamus was detected byELISA kits. The mRNA level of hippocampus NR2A glutamate receptor was detected byreal-time quantitative PCR. The results showed that significant difference of appearance wasfound in sub-health model mice compared with normal mice, and the behavioral data statisticswere lower than normal group(P<0.05). It suggested that using the compound factors to establishsub-health mice was successful. After administration with Lycium barbarum polysaccharide for21days, the blood routine, blood biochemistry and pathological examination were not abnormalin experimental mice. The duration of sleep in Lycium barbarum polysaccharide groups wassignificantly longer than that in model group and the normal group (P<0.01). The activity ofSOD and CAT was lower and MDA level was higher in model groups than those in normal group(P<0.01). However, the activity of SOD and CAT was increased and MDA level was decreasedin high dose Lycium barbarum polysaccharide group. The thymus index, spleen index, B cellproliferation ability and CD4+/CD8+ratio in model group were lower than those in normal group,however, these indicators were improved after Lycium barbarum polysaccharide treatment for4w. The DA content in hypothalamus and NR2A mRNA expression level in hippocampus ofmodel group were all lower compared with control group, however,5-HT content of model group was higher than normal group. After the administration of Lycium barbarum.polysaccharide, the DA content and NR2A mRNA expression level in high dose group weresignificantly increased (P<0.05), and5-HT content reduced observably (P<0.01). In a word,Lycium barbarum. polysaccharide improved the sub-health state of mice, which associated withmeliorating sleep, antioxidation, enhancing immunity and antifatigue.
