Crude lycium barbarum polysaccharides(LBP) was extracted from Ningxia-lycium barbarum by hot-water extraction technique. And the content of saccharide was measured by sulfuric acid-phenol method; The animal model of non-insulin-dependent diabetis mellitus(NIDDM) was estabilshed by high-energy feeding combined with the injection of small-dose streptozotocin(STZ). According to the area under the curve of glucose tolerance, the level of insulin(Ins) and the insulin sensitivity index(ISI), the rats of NIDDM model were divided into four groups, including model group, LBP low dose group, LBP middle dose group, and LBP high dose group. At the same time, normal rats were divided into normal group and LBP high dose group. The changes of the levels of blood glucose, lipid, insulin, advanced glycosylation endproducts(AGEs), SOD, MDA and creatinine(CR)in serum and the liver glycogen, CR in urine, hemoglobin-A1c(HbA1c) in plasma, aldose reductase(AR) in kidney, IL-8 of renal cortex were detected. The pathology of kidney and pancreas was observed in order to research the effect of LBP on reducing blood glucose and preventing the occurrence of diabetic nephropathy(DN) and to explore the mechanism of this effect. The following is the results:A. The average extraction rate of crude LBP was 7.4%, after the Lycium Bararum was treated by the following procedure: baking, defatting, discoloration, removing small molecular sugar, deposition by ethanol, cleaned by organic solvent and vacuum drying. The average content of saccharide was 45%(detected by sulfuric acid- phenol method).B. The crude LBP obviously delayed the weight decreasing of rats, reduced the 24h-urine volume, the levels of glucose, AGEs-P in serum, HbAlc in plasma, IL-8 in renal cortex and AR in kidney. At the same time, LBP increased the level of liver glycogen, insulin in serum, ISI of the model rats, and the action of SOD in serum, but the crude LBP has no effect on lipid content in serum. In addition, the crude LBP increased the endogenous creatinine clearance(ccr) of the model rats. The crude LBP has significant effect of reducing the level of glucose in serum and protecting the renal function of NIDDM model rats.
