Objectives:To evaluate the cytological features of the two kinds of electrospunRana Skin Collage/PLLA nanofibrous composite, electrspun in differenttextile technologies, from the cytological experiments in different ways.Methods:The human skin fibroblasts were seeded on the electrospun Rana SkinCollage/PLLA nanofibrous memberanes which were fixed on the glassslides. Using the Cell Counting Kit to test the cell proliferation andcytotoxicity of nanofiber membranes. A scanning electron microscope wasprepared to observe the cell morphology and adherence to the nanofibermembranes. Observing the cell growing patterns on the nanofibermembranes by laser scanning confocal microscope.Results:No cytotoxicity of the electrospun Rana Skin Collage/PLLAnanofibrous composite was proved using the Cell Counting Kit,but the cellproliferation speed was found different on the two kinds of nanofibermembranes, the cells of the oriented group was growing slower than theones of the random-aligned group. A well cell adherence was found on thenanofiber membranes, and we preliminarily discovered that the cellsseeded on the oriented nanofiber membranes were proliferating faster thanthose seeded on the random-aligned nanofiber menbranes. We used laser scanning confocal microscope to find that the cells of the oriented groupsgrew aligned according to the long axis of nanofibers while the others grewin a disordered way.Conclusions:The random-aligned electrospun Rana Skin Collage/PLLAnanofibrous composite seemed more suitable for the the human skinfibroblasts’ growth characteristics, on which a higher rate of cellproliferation was showed with statistical significance. The electrospunRana Skin Collage/PLLA nanofibrous membrane is a new kind ofnanofibrous composite which may be capable of inducing the cellproliferation and promote the wound healing...
