| Objective:The aim of this study was to investigate the effect of Bioactive Peptide from the Skin of Rana Chensinensis on immune function in normal mice.Method:The mice were randomly divided into five groups as follow:Normal control groups(NG:with Distilled water), Low-dosage groups (LG:100 mg/kg), Middle-dosage groups (MG:500 mg/kg), High-dosage groups (HG:1000 mg/kg) and positive control groups (PG:1000 mg/kg)with VC. Every group was assigned ten mice, which should be arranged to be weighted one time every three days, the groups received by gastric perfusion once every day. Then according to weight, we administrated the dosage of gastric perfusion for 28 days.24 hours after the last time were given medicine, the mice were sacrificed by cervical dislocation, then we did the experiment of mice' viscera index,ConA-induded mice splenic lymphocyte transformation(MTT), SRBC-induced tardive allergic reaction(Foot metatarsal thickening method), antibody generating capacity test(Jerne), serum hemolysin level test(haemagglutination test), carbon clearance test, test of phagocytosis of peritoneal macrophages(in situ Experiment) and test for determination of activity of natural killer cell for oberserving various result.Result:1,Thymus index could be improved in HG and PG, there was significant difference between HG,PG and NG. (P<0.05). Spleen index could be improved in HG and PG, there was significant difference between HG, PG and NG (P<0.01).Improving of viscera index of HG was better than PG. There was significant difference between HG and PG (P<0.05).There was no signifficant difference between LG, MG and NG (P>0.05)2,Both SI index of MG and HG were improved. There was significant difference betweenMG, HG and NG (P<0.01). There was no significant difference between LG and NG (P>0.05). There was signifficant difference between PG and NG (P<0.05).Improving of SI index of MG and HG was better than PG. There was significance difference between MG, HG and PG (P<0.05)3,DHT of mice can be induced to higher in HG. There was significant difference between HG and NG (P<0.01. DHT of mice can be induced to higher in PG. There was significant difference between PG and NG (P<0.05). Induction of HG was better than PG. There was significant difference between HG and PG (P<0.05). There was no significant difference between LG,MG and NG (P>0.05)4,LG, MG, HG and PG can enhance the Antibody formation of mice, they are very difference from NG(P<0.01). The generation of antibody of MG was better than LG and HG. There wsa significant difference between MG and LG, HG (P<0.05)5,The hemolysis value of HG and PG was improved. There was significant difference between HG,PG and NG (P<0.01). The hemolysis value of LG and MG was improved. There was no significant difference between LG, MG and NG (P>0.05)6,On carbon clearance ability test, Macrophage phagocytic index can be enhanced in HG and PG. There was significant difference between HG,PG and NG (P<0.01). There was no significant difference between LG, MG and NG (P>0.05)7,On phagocytes test of mouse abdominal cavity phagocyte with the chicken erythrocyte, Phagocytic rate and Macrophage phagocytic index can be enhanced in LG, MG, HG and PG. There is significant difference between LG, MG, HG, PG and NG (P<0.01). The effect of HG was best in all of all groups. There was significant difference between HG and LG, MG (P<0.01). There was significance difference between HG and PG (P<0.05)8,Nk cell's activity of HG, MG, PG was improved. There was significance difference between HG, MG, PG and NG (P<0.05). The effect of HG was better than MG and PG, There was significance difference between HG and MG, PG (P<0.05)Conclusion:The result indicated that Bioactive Peptide from the Skin of Rana Chensinensis could enhance cellular immunity, humoral immunity and non-specific immunity of mice, thus improving the capacity of organism on resisting harmful factors which were the outside invasion. This study can provide scientific evidence for Skin of Rana Chensinensis' comprehensive utilization in health food and medical area. |
