ObjectiveCompared the different functions of neovasculization on human microvascular endothelial cell (HMEC-1) between Xuefu Zhuyu Decoction and VEGF. To establish the foundation of the further study about the differences of regulation mechanisms of angiogenese between Xuefu Zhuyu Decoction and VEGF. And then provide the modern scientific evidence of the clinical application of Xuefu Zhuyu Decoction, and to avoid the side effect of VEGF in the treatment of promoting neovasculization.MethodCollected and separated the healthy volunteer’s serum who took five dose of Xuefu Zhuyu Decoction after7days, and the healthy volunteer’s normal serum. Divided the human microvascular endothelial cell (HMEC-1) randomly which cultured in routine into11groups. Set VEGF dose of1.25ng/ml、2.5ng/ml、5ng/ml、10ng/ml, and0ng/ml asVEGF interfering group and blank group. Set the dose of1.25%、2.5%、5%, et al, as the blank control and the Tradition Chinese Medicine interfering group. Dissolved each drug in MCDB-131culture media contained5%FBS, and cultured for24h、48h and72h. Observed the influence of the different drugs had on the reduplication, immigration, adhesion, microvascular by the means of MTT colorimetric technique, cell cycle test, Boyden chamber, adhesion test and the test of vascular neogenesis in vitro test in vitro.Results1. The different density of serum can affect the activity of cell proliferation, migration, adhesion, neovasculization, so it may be objective to make the four concentration of VEGF compared with blank group, the same amount of the Xuefu Zhuyu Decoction-contained serum and control serum group respective.2. Each concentration of VEGF can significantly promote cell activity earlier than the Xuefu Zhuyu Decoction.3. To the influence of cell migration, VEGF and the Xuefu Zhuyu Decoction both can significantly improve cell migration, but VEGF had slower effects with cells.It showed a greater role in promoting cell migration, until affected48h and affected with time dependence.4. In the aspect of promoting cell proliferation, each concentration of VEGF and Xuefu Zhuyu Decoction showed continued effect on cells.5. All concentration VEGF can promote cell proliferation at the three time points, and had the time dependence. While the high concentration of the Xuefu Zhuyu Decoction-contained serum showing the inhibition at the72h time points.6. All concentration VEGF can promote vascularization at the three time points.,the effect is longer than the Xuefu Zhuyu Decoction.And only the lower concentration of the Xuefu Zhuyu Decoction can promote cell vascularization in short time.Conclusion1. Both Xuefu Zhuyu Decoction and VEGF can significantly promote the HMEC-1cell neovasculization.Their mechanism may relative to promoting cell proliferation, migration and adhesion.2. The two drugs both had the same trend in promoting cell adhesion.While had differences about the effect on cell proliferation, migration, and vascular neogenesis. In different time points each concentration of VEGF can promote vascularization.and the time may be longer than Xuefu Zhuyu Decoction. In some conditions, the Xuefu Zhuyu Decoction showed inhibitory effect on the cell, and it showed a short time to promote vascularization.It is the cue that Xuefu Zhuyu Decoction may regulate the angiogenesis, and different with the VEGF, that promoted vascularization continued.
