Objective:Effect of Xuefu Zhuyu Decoction on the concentration of TXA2and PGI2in plasma and anticoagulant and fibrinolytic System of Arterial Thrombosis Model in Rats.Methods:Choose60SD rats, half male and half female, were randomly divided into blank group, model group, Aspirin group, Xuefu Zhuyu Decoction of high, medium and low dose group. All Rats were giving prophylactic intragastric infusion once a day for fourteen continuous days, During this experiment, Xuefu Zhuyu Decoction of high, medium and low dose groups were respectively received13.68,6.84,3.42g/(kg.d). Aspirin control group was received aspirin orally10mg/(kg.d).model group and blank group were received equal volume of physiological saline.Weighing the weight of rats two times a week and according to the change to adjust the gavage dose at any time.then fecl3was applied onto the carotid artery to induce arterial thrombosis except the blank group. Ambilateral common carotid artery was separated and the right common carotid artery was used for sampling blood. A piece of plastic film(1cm×1cm) with the solution20uL35percent FeCl3was put under the3cm long part of the separated left common carotid artery for 30min,After the piece of filter paper removed, blood was from the common carotid and3000rpm|min for15minutes at4℃,and the concentration of TXA2, PGI2, t-PA, PAI-1, PLG, AT-III and PC in plasma were determined by enzyme linked immunosorbent.Then the thrombotic segement of the vessel was taken and weighted.Result:1.35%ferric chloride can make the carotid artery of model group rats has obvious thrombosis,, Indicating a successful model.2. Comared with the model group, the thrombus weight were lighter in drug groups and the differences were significant(p<0.01),In which the thrombus weight lightest was medium dose group (p<0.01);3. Compared with blank group, the concentrations of TXB2, PAI-1, the ratio of TXB2and6-keto-PGF1a in plasma were significantly higher in model group,while the concentrations of6-keto-PGF1a, t-PA, PLG, AT-Ⅲ, PC, the ratio of t-PA and PAI-1were significantly lower in model group (p<0.01);4. Compared with the model group, the concentration of TXB2was significantly decreased and6-keto-PGF1a in plasma was significantly increased in different doses Xuefu Zhuyu Decoction group,the differences were significant (p<0.01);5. Compared with the model group, The concentration of t-PA and t-PA/PAI-1in plasma were significantly increased in Xuefu Zhuyu Decoction of high and medium group, while the concentration of PAI-1and PLG in plasma were decreased,the differences were significant (p<0.01);6. Compared with the model group, The concentration of AT-Ⅲin plasma was significantly lower in different doses Xuefu Zhuyu Decoction group, while the concentration of PC was significantly increased (p<0.01);7. Aspirin has obvious anti-thrombotic effect, but it did not influence the content of AT-IIIand PC in the plasma. It also did not influence t-PA and PAI-1content in the plasm. while the concentration of TXB2was significantly decreased and6-keto-PGF1a in plasma was significantly increased in Aspirin group(p<0.01),Conclusion:1. Xuefu Zhuyu Decoction can prevent vascular endothelium from injury and increase the anti-thrombosis function of endothelial cells.2. Xuefu Zhuyu Decoction by increasing the concentration of6-keto-PGF1a and reducing the concentration of TXB2in plasma of rats,the antithrombotic mechanisms of Xuefu Zhuyu Decoction may be brought by restraining platelet aggregating to inhibit thrombosis.3. Antithrombotic mechanisms of Xuefu Zhuyu Decoction may be related to binding with AT-Ⅲ,and it could also prevent the consumption of PC after carotid arterial thrombosis. it may concern inactivating coagulation factors by binding with AT-Ⅲ or accelerating inactivation of coagulation factors by PC. the action of its mechanism may have correlation with can increase the activity of the anticoagulant system.4. Xuefu Zhuyu Decoction by increasing the concentration of t-PA and reducing the concentration of PAI-land PLG in plasma of rats,By adjusting the ratio of t-PA/PAI-1to achieve the correct balance between them.the action of mechanism may have correlation with increasing of the activity of the fibrinolysis system.
