The Effect Of EphrinB2/EphB4in Angiogenic Mechanisms Of Xuefu Zhuyu Decoction

ObjectiveStudy the relation between ephrinB2and its receptor EphB4to Xuefu Zhuyu Decoction(XFZY) in angiogenic activities by using human umbilical vein endothelial cell-12(HUVEC-12), and make a base for further research on XFZY of its angiogenic mechanism.Method1. Cultured HUVEC-12with10%fetal bovine serum (FBS) and5%fetal bovine serum separately.2. By making mouse Xuefu Zhuyu Decoction-Contained Serum(XZDCS), incubating the cells with5%、2.5%and1.25%XZDCS seperately for24h、48h and72h, and then examing their proliferation、migration、adhesion and target genes expression activities by the means of MTT colorimetric technique、Boyden chamber、adhesion test and RT-PCR (Reverse Transcription-Polymerase Chain Reaction method).Result1. In the aspect of proliferation, different concentrations of XZDCS play significant promoting effects on24h and48h (P<0.05or P<0.01).2. In the aspects of migration and adhesion, all of three concentrations play significant promoting effects on24h、48h and72h (migration P<0.05or P<0.01, adhesion P<0.01).3. In the aspect of ephrinB2expression, some concentrations play promoting roles on all three time points (P<0.05or P<0.01).4. In the aspect of EphB4expression, some concentrations play promoting roles on24h and72h time points (P<0.05or P<0.01), particularly on72h (P<0.01).Conclusion 1. XFZY have significant angiogenic effects,which closely related to proliferation、migration、 adhesion activities of HUVEC-12.2. Some cocentrations of XFZY can significantly promote ephrinB2expression on24h、48h and72h, and could also promote EphB4expression on24h and72h.
