| Objectives:This study is to observe the number of Expression of LC3and Beclinl in the per-ihematoma brain region of intracerebral hemorrhage (ICH) patients by using Immunohist-ochemistry and imaging analysis, so as to discuss the relationship between expression and Sthenia ZHENG, Protration ZHENG, volume of hematoma, scores of NIHSS and GCS. To investigate the effect of autophagy and the possible mechanisms in secondary injury after intracerebral hemorrhage. To provide the molecular biology standard for the microcosmic syndrome differentiation with traditional Chinese medicine (TCM).Materials and methods:26patients of ICH who were collected during the surgical operation with the diagnosis of ICH and the sthenia and prostration ZHENGs of stroke were observed.Among them, there were13male and13females, the average ages were56(34-83years old);11were yang sthenia,among them6males and5female,average ages were53(38-72years old);10were yin sthenia,among them3males and7females,average ages were64(39-83years old);5were prostration,among them4males and1female,average ages were49(34-65years old);collected the brain tissues which was drained during the operation of clear up haematoma.6normal subjects as control which are normal tissues. The DUO TIAN formula was used to calculate the volume of hematoma, the NIHSS and GCS was used to score. The specimens are made strictly by the related methods. We observe the expression of LC3and Beclinl in the peri-hematomal brain region by using immunohistochemistry and imaging analysis.Results:1.The number of the expression of LC3and Beclinl are significantly increased in the peri-hematomal brain region of ICH Patients compared with that in the normal control group(P<0.05).2.The number of the expression of LC3and Beclinl are significantly different among YANG Sthenia ZHENG,YIN Sthenia ZHENG and Protration ZHENG group(P<0.01). The number of the expression of LC3and Beclinl:the first one is Protration ZHENG group, the second one is YIN Sthenia ZHENG and the last one is YANG Sthenia ZHENG3.The number of the expression of LC3and Beclinl display a positive role to the volume of hematoma in the peri-hematomal brain region of ICH Patients(P<0.01).4.The number of the expression of LC3and Beclinl display a positive role to scores of NIHSS,but the number of the expression of LC3and Beclinldisplay a negative role to scores of GCS(P<0.01).5.The volume of hematoma and scores of NIHSS and GCS are clearly different among YANG Sthenia ZHENG,YIN Sthenia ZHENG and Protration ZHENG group(P<0.01).The volume of hematoma and scores of NIHSS:the first one is Protration ZHENG group, the second one is YIN Sthenia ZHENG and the last one is YANG Sthenia ZHENG, the scores of GCS is adverse.Conclusion:1.There is few of the expression of LC3and Beclinl in the normal control group by using immunohistochemistry and imaging analysis. Lysosomal autophagy pathway can be activated in the peri-hematomal brain region of ICH Patients,there is more expression of LC3and Beclinl in the nerve cells.2.In the peri-hematomal brain region of ICH Patients, the number of the expression of LC3and Beclinl display a positive role to the volume of hematoma and scores of NIHSS, the number of the expression of LC3and Beclinl display a negative role to scores of GCS.So we speculate that autophagy may be involved in the secondary brain injury after ICH in human.3.The expression of the expression of LC3and Beclinl in the peri-hematomal brain region of ICH Patients can provide some help for the microcosmic syndrome differentiation with TCM.Combining with the volume of hematoma,scores of NIHSS and GCS maybe provide evidence on diagnosing Sthenia ZHENG and Protration ZHENG with TCM. |
