[Objective]This paper aims at observing the influence of general situation, ovaries, uterus index and histomorphology, female progestational hormone and its receptor expression with kidney deficiency and blood stasis syndrome treated by assisted reproduction1prescription and exploring function of assisted reproductionl prescription which could improve ovarian,uterus tissue morphology and the endocrine,as well as analyzing its possible mechanism, by which it can provide theoretical support in clinical application of assisted reproduction.[Method]Mature SD50unfertilized female rats, raising a week later, on the basis of prooestrus, rats were randomly divided into blank group, model group, high and low dose group, wu zi yan zong pill group, taking wu zi yan zong pill group as positive drug control group. Model group, low dose group, high dose group and positive control group use hydroxyurea to establish model of kidney deficiency, on the eight day, discontinued hydroxyurea. Line the vagina pictures, in estrus in cage, in the4days of pregnancy, except the blank group and positive control group, the rest of the groups use high molecular dextran to establish model of blood stasis.after thirty minutes,the rats were killed,and taken out rats uterus and ovaries, each group of randomly selected6samples. After HE staining, we observed endometrium and ovarian tissue under the microscope.reflection immune analysis method is used to inspect the blood estradiol, progesterone, using immunohistochemical method to detect the endometrium of female, progestational hormone receptor expression.[Result]1. The influence of weight gain of model rats with kidney deficiency and blood stasis syndrome by assisted reproduction1prescription.Compared with the blank group, model group rats weight gain is small, the difference was statistically significant (P<0.05);Compared with model group, the high and low dose of assisted reproduction1prescription group rats weight gain are obvious, the difference were statistically difference (P<0.05).2.The influence of ovaries index and viscera index of model rats with kidney deficiency and blood stasis syndrome by assisted reproduction1prescription.Compared with the blank group, model group rats index of uterus and ovarian are low, differences were statistically significant (P<0.01); Compared with model group, assisted reproduction1prescription of high and low dose groups can promote the growth of rat uterine viscera index, the difference was statistically (P<0.05), assisted reproduction1prescription of high and low dose group can promote the growth of rat’s uterine and ovaries viscera index,but there were no statistically significant difference (P>0.05).3.The influence on ovarian tissue morphology with kidney deficiency and blood stasis syndrome by assisted reproductionl prescription.Compared with the blank group, model group overall follicle and corpus luteum number are less, differences were statistically significant (p<0.05);Compared with model group, assisted reproduction1prescription high and low dose groups luteum number are more, the difference was statistically significant (p<0.05), the total number of follicles at different levels and mature follicle Numbers are more than the model group, but there was no statistically significant difference (p>0.05).4.The influence on serum estradiol (E2) and progesterone (P) with kidney deficiency and blood stasis syndrome by assisted reproduction1prescription.Compared with the blank group, estradol and progestogen of model group rats are lower, differences were statistically significant (P<0.01); Compared with model group, assisted reproduction1prescription of high and low dose groups of estradol and progestogen are higher, the difference was statistically significant (P<0.05)5.The influence on endometrial morphology with kidney deficiency and blood stasis syndrome treated by assisted reproductionl prescriptionEndometrial glands of model group rats are scattered, scarce, narrow and straight cavity, stroma were loosened, major of cells were bipolar spindle shape, small spiral arteries are located in deep of intima layer, and assisted reproduction1 prescription of high and low dose groups endometrium of rats with different levels of development, glandular cavity is larger, part under a nuclear vacuoles, nuclear egg circle, nucleoli.Interstitial edema, the cells showed ovoid or star, nuclear egg circle, helical, blood vessels and close to the function layer, wall thickening, approximation with the blank group.6.The influence on estrogen receptor (ER) and progesterone receptor (PR) expression of endometrial tissue with kidney deficiency and blood stasis syndrome treated by assisted reproductionl prescriptionCompared with the blank group, model group the positive expression of endometrial ER and PR is less, the difference was statistically significant (P <0.05).Compared with model group, assisted reproduction1prescription high and low dose groups of ER and PR in endometrium positive expression have different extent increase, the difference was statistically significant (P<0.05).[Conclusion]1.Assisted reproduction1prescription could improve the effect of rats of renal deficiency and blood stasis type, which can be found that expression in not only the improving of spirit, activity, diet and the color of the fur, defecate, claws, the tail and ears but the improving of decreasing weight.2.Assisted reproduction1prescription could lead developing of endometrium to a benefit of embryos to attach to the uterus by improving development of ovary and uterus of rats, promoting follicular maturation and luteinization, increasing the ovarian endocrine functions, balancing the level of E2and P.3.Assisted reproduction1prescription can provide a favorable endometrial environment for the blastocyst embedding by controlling the expression of ER and PR in endometrium.
