Objective: Lung cancer is one of the major malignant tumors harming people’s healthin recent years. Up till now,lung cancer treatment seems to have not reached a significantbreakthrough besides targeted therapy in advanced and metastatic. Therefore, it isparticularly important to investigate the mechanism of invasion and metastasis of lungcancer and to provide new thoughts for the management of lung cancer and improve thesurvival rate. A recent study suggested that Uroplakin1A down-regulation or deletion playan important role in the process of the invasion in lung cancer,MMP7was found highlyexpressed in lung cancer if Uroplakin1A was found down-regulation or deletion,andCyclin D1amplification rate is much higher in lung squamous cell carcinoma than othernon-small cell lung cancer. Therefore, we examined the expression of Uroplakin1A,Cyclin D1and MMP7in non small cell lung cancer tissue and to investigate itscorrelation.Methods: Paraffin-embeded lung cancer specimens of126patients from2011to2013were prepared in the pathological department of our hospital, including66cases oflung squamous cancer and60cases of lung adenocarcinoma cancer;60cases of lungtissues adjacent to cancer were also gathered, adjacent tissue of lung squamous cancer was30cases, the same number with the adjacent tissue of lung adenocarcinoma cancer.Thetotal of186cases of blocks were4~6μm sectioned, immunohistochemistry (ICH) wasused to detect the expression of protein of Uroplakin1A, MMP7and Cyclin D1in thosetissue samples, Correlation between the expression of those proteins and clinicalpathological differentiation, tumor size, TNM stage were analysed, Correlation ofUroplakin1A, MMP7and Cyclin D1were also analysed in order to investigate theexpression and significance of Uroplakin1A, MMP7and Cyclin D1in lung cancer. At thesame time, Western blot was used to detect the protein expression with Uroplakin1A,MMP-7, Cyclin D1in Non Small Cell Lung Cancer Series including large cell lung cancercell lines (H460), lung adenocarcinoma(A549, H1650) and lung squamous cell carcinoma (H520, H226),in order to investigate the expression relationship of the protein in differenttypes of cell lines.Results: Western blot examination revealed: MMP7and Cyclin D1protein werehighly showed in large cell carcinoma, adenocarcinoma and squamous cell carcinoma, butH520and H226lung squamous cell carcinoma cell lines with low Uroplakin1A expression;Proteins of Uroplakin1A, MMP7and Cyclin D1expression were detected byimmunohistochemistry in Non Small Cell Lung Cancer and their adjacent tissues, andobserved the following phenomenon:1) positive rate of Uroplakin1A in lung squamouscell icarcinoma decreased obviously compared with its adjacent tissues (48.48%,96.67%),but lung adenocarcinoma and its adjacent tissues showed high positive expression rate(75%,90.00%).2) Positive rates of MMP7in lung squamous cell cancer and lungadenocarcinoma cancer were high (46.97,70.00%),but low in the adjacent tissues tissues(6.67%,16.67%). This phenomenon was same with the positive rates of Cyclin D1in lungsquamous cell cancer and lung adenocarcinoma cancer (31.82%,38.33%) and theiradjacent tissues tissues (3.33%,16.67%).3) The positive rate of Uroplakin1A wasdecreased in advanced stage and poor differentiation compared with early stage andwell-differentiation in lung squamous cell cancer, while the positive rates of MMP7andCyclin D1were opposited in such situation.4) The expression of Uroplakin1A has anintensity correlation with MMP7and Cyclin D1in lung squamous cell cancer, correlationcoefficient were0.344and0.320, P value respectively0.003and0.006.5) The expression ofUroplakin1A was correlated with stage, differentiation and tumor size, P value was lessthan0.05Conclusion: The expression of Uroplakin1A protein was low in lung squamous cancercell line, Uroplakin1A was low expression in lung squamous cell carcinoma but high inlung adenocarcinoma cancer and lung cancer adjacent tissues, Cyclin D1and MMP7protein were high expression in lung cancer but low in its adjacent tissues, Uroplakin1Awas related with tumor staging, differentiation degree, tumor size,Cyclin D1and MMP7inlung squamous cancer.
