Cortex phellodendri is one of mark herbs, first recorded in Shengnong ben cao jing, as a top grade.With the medical history of more than two thousand years. Cortex phellodendri bitter and cold, it has the role of clear heat and dry dampness, purging fire for removing toxin. Supported by Study on grading specifications and quality evaluation of Phellodendron Chinense and its Pieces (201007012-3-2) in State Chinese Medicine Administration Bureau, we have divided Cortex phellodendri in to two grades, then formulated the relevant quality standards.OBJECTIVEThis experiment is aimed at the improving of quality standards of different specifications and different processed products of Cortex phellodendrine, thus established a quick and stable method to evaluate the quality of cortex phellodendri, so we could control the quality of cortex phellodendri better.METHODS1To establish a new method which quantitative analysis of multi-components of five alkaloids in Cortex Phellodendri cortex. Berberine, the main component in Cortex Phellodendri, was selected as the internal reference substance, relative correction factor(RCF) between berberine and magnoflorine, phellodendr-ine,palmatine as well as jatrorrhizine were established. Comparied quantitative results the method of external standard and QAMS of the five contents in30batches of samples.2To collect NIR diffuse reflectance spectra of samples, regard alkaloid measured by HPLC as reference, then using partial least square algorithm (PLS) established a calibration model, comparison the influence of different pretreatment method and different wave for modeling.A near-infrared(NIR) quantitative method was developed for rapid determination of three components (berberine, phellodendrine, magnoflorine) in Cortex Phellodendri.3By using new methods to evaluated inner quality in different specifications and different processed products of Cortex phellodendrine.RESULTS1No significant differences were found in the quantitative results of five alkaloids in30batches of Cortex Phellodendri determined by external standard method and QAMS (RSD<5%).2Have established the the identification model of cortex phellodendri samples, different regions of cortex phellodendri and different processed products cortex phellodendri.Take MSC+2D2as pretreatment method, in4342.91-5006.3,5804.68-7500,8000-10000band, R2for berberine, phellodendrine, magnoflorine were0.9995,0.9760,0.9306,0.9995,RMSEC for berberine, phellodendrine, magnoflorine and total alkaloids were0.0453,0.0335,0.0396,0.0498.CONCLUSIONThe study use a combination of muiti-index quality control and NIR inner quality in different specifications and different processed products of Cortex phellodendrine. It is reliable and accurate to evaluate the quality of Cortex phellodendrine.
