The dry bark of Cortex Phellodendri of rutaceae plants trees Phellodendron chinense Schneid. was known as Sichuan Cortex Phellodendri. Alkaloids were the main active ingredients in Cortex Phellodendri, and berberine possesses the highest content among those alkaloids. Berberine exhibits various biological activities, such as antibacterial, antiviral, antitussive, depressurization and enhance immunity and so on and is getting more and more attention. Based on the ACCE new continuous counter-current extractor, a continuous countercurrent extraction tests in laboratory simulation scheme was established to extract berberine in Cortex Phellodendri. The final aim is to conduct a comprehensive and systematic process to develop a set of simple, environmental protection, low cost, and large scale preparation of high purified berberine. This experiment has achieved the following progress:1. This study established HPLC/DAD detection method to analyze berberine and phellode ndrine at the same time. It laid a basis for efficient and reasonable utilization of young cortex phellodendri raw material by comparing the content and biomass of berberine of c ortex phellodendri in different growth duration and different harvest time.2. This study set up a simulation scheme of continuous counter-current extraction in labo ratory. and provided a laboratory simulation method. The experiment study provided a re search idea and design simulation steps of ACCE continuous counter-current test in labor atory. In addition, we studied various parameters for get the theoretical maximum concen tration in extraction production.3. The berberine extract liquid mass fraction up to90.5%was achieved by using ACCE continuous counter-current extraction equipment.4. Finally, we separated and purified obaculactone from cortex phellodendri.and the mass fraction of Obaculactone reached91.99%. This technology could contribute to explore a feasible scheme for industrialized isolation and purification of wastes residue after the c ontinuous counter-current extraction in the factory.
