Objective:Gambogic acid shows anti-inflammatory effect in many studies. But the effect of gambogic acid on articular cartilage metabolism remains unknown. In this study, we investigate the protective effect of the gambogic acid (GA) on OA.Methods:For the in vitro work, rabbit articular chondrocytes were cultured, and quantitative real-time polymerase chain reaction (qRT-PCR) was used to investigate changes in gene expression upon the addition of GA in chondrocytes processed with IL-1β, changes in protein profiles were assessed by Western blotting. In vivo study, a rabbit anterior cruciate ligament transaction model of OA was established. The rabbits were given intra-articular injections of GA.Results:GA inhibited the expression of Matrix Metallo Proteinases(MMP)-3,-13and increase the expression of TIMP-1, SHH in IL-1β-induced rabbit chondrocytes in vitro work. And in vivo study, GA showed the same effect of suppressing cartilage degradation.Conclusion:GA play a protective role in the development of OA and may be helpful in the treatment of OA. |
