Abstract/Summary:
ObjectiveDN is the one chief pathoqeny of ESRD, therefore it has important meaning to explor machnism and preventive treatment of DN. a series of data were evaluated such as the ratio of kidney weight to body weight,serum glucose,renal function,24h microalbuminuria of rats with diabetes; research of leech on the expression of MMP-9, TIMP-1,TGF-β1 in kidney by immunohistochemical stain,the express of IV-C and TGF-β1 by euzymelinked immunosorbent assay(ELISA). observe structure of kidney under by light microscope and transmission eletron microscope in kidney of rats with diabetes; to study the effect and mechanism of leech on early kidney disease of diabetes nephropathy rat.Methods45 clean level SD rats were randomly divided into normal control group, model group, Teech treatment group, control group. DN rat model were injected STZ (55mg/kg) to induce diabetic rat model, group C:diabetic nephropathy rat received daily intragatric administration of Leech at dose of 1g/kg/d; group D:DN rat received daily intragatric administration of Telmisartan at dose of 30mg/kg/d; group A and B:normal rat and DN rat received daily intragatric administration of distilled water. All group were sacrificed after eight weeks. Results①biochemistry index:compared with the group A, ratio of kidney weight to body weight,serum- glucose,renalfunction,24hmicroalbumin increased in group B,C and D (P<0.01), compared with the group B, ratio of kidney weight to body weight, serum glucose, renalfunction,24h microalbumin decreased in group C and D (P<0.01),②WE observed mesangial proliferation was obvious in group B and it reduced in C and D group as compared with B by light microscope. foot procexs fusion, widened mesangium in group B by transmission eletron microscope and a better improvement in C and D group.③immunization index:compared with the group A, the expression of TGF-β1,TIMP-1 increased in group B,C and D(P<0.01), the expression of MMP-9 decreased in group B,C and D(P<0.01), compared with the group B, the expression of TGF-β1,TIMP-1 decreased in group C and D (P<0.01), the expression of MMP-9 increased in group C and D(P<0.01).③ELISA index:compared with the group A, TGF-β1 andⅣ-C of serum increased in group B,C and D(P<0.01), compared with the group B, TGF-β1 andⅣ-C of serum decreased in group B,C and D(P<0.01).④correlation analysis:TGF-β1 and MMP-9 is negative correlation r=-0.750(P<0.01); TGF-β1 and TIMP-1 is positive correlation r=0.891 (P<0.01) MMP-9 and TIMP-1 is negative correlation r=-0.820 (P<0.01); uAlb and blood TGF-β1 is negative correlation r=0.882 (P<0.01); uAlb and bloodⅣ-C is positive correlation r=0.928 (P<0.01)Conclusion:The expression of MMP-9, TIMP-1, TGF-β1 and blood serum IV-C are related to DN, leech has protective effect on DN through affecting the chang of above mentioned indexes.
