PartⅠExpression and clinical manifestation of matrix metallo proteinases ingastric adenocarcinomaObjective: To study the relationship between clinical manifestation of gastricadenocarcinoma and the expression of matrix metalloproteinases 2 protein (MMP-2),matrix metalloproteinases 9 protein(MMP-9). Methods: Total 83 patients with gastricadenocarcinoma were included to determine the expression of the MMP-2,MMP-9 in thesample from surgery, using immunohistochemical staining. Two independent pathologistswere enrolled for the evaluation of the staining. Results: The positive rate of staining forMMP-2,MMP-9 were 72.29%,63.86% respectively. An positive correlation wereobserved between MMP-2, MMP9 expression and TNM classification(P<0.05) andnode metastases(P<0.05), furthermore, MMP-9 had an correlation with the deep of cancerinvasive. Conclusions: MMP-2 and MMP-9 expression in the cancer were related to theaggressiveness of the gastric cancer and might serve as malign factors for prognosis, thisexperiment uses immunohistochemistry method to study matrix metalloproteinases ingastric adenocarcinoma, It has important referring value that on clinical judges the tumorshift possibility, the biology behavior and the instruction later period clinical treatment.PartⅡExpression and clinical manifestation of tissue inhibitors of matrixmetalloproteinases in gastric adenocarcinomaObjective: To study the relationship between clinical manifestation of gastricadenocarcinoma and the expression of tissue inhibitor of matrix metalloproteinase 1 protein (TIMP-1) and tissue inhibitor of matrix metalloproteinase 2 protein. Methods:Total 83 patients with gastric adenocarcinoma were included to determine the expressionof the TIMP-1,TIMP-2 in the sample from surgery, using immunohistochemical staining.Two independent pathologists were enrolled for the evaluation of the staining. Results:The positive rate of staining for TIMP-1,TIMP-2 were 72.29%,63.86% respectively.The expression of TIMP-1 the gastric carcinoma has significance (P=0.001) in two groupof different degrees, also its expression is good in the high and medium degree group, butis bad in the low degree group, it presents the correlation in the degree of differentiation.The expression of TIMP-2 the gastric carcinoma has significance (P=0.008) in two groupof different degrees, also its expression is good in the high and medium degree group, butis bad in the low degree group, it presents the correlation in the degree of differentiation.An positive correlation were observed between the expressions of TIMP-1 and TIMP-2 inthe gastric cancer tissue were more prevalent in the patients with low TNM classification,small size of cancer, highly differentiation or without node metastases. The correlationanalysis indicates expression of TIMP-2 in gastric carcinoma is negative correlation withexpression of MMP-2(r=0.364,p=0.01). But the correlation of TIMP-1 and MMP-2,TIMP-1, TIMP-2 and the MMP-9 is not significant different. Conclusions: TIMP-1 andTIMP-2 expression might be the protective factors for the gastric cancer. This researchindicated that existed the relevance between the increase of MMPs and the decrease ofexpression of TIMPs. The maning of the only the increases of the expression of MMPs orthe decreases of the expression of TIMPs is inferior to the imbalance of MMPs/TIMPs byfar.the presence of TIMP-1 and TIMP-2 in gastric carcinoma had extensive heterogeneity.TIMP-1 and TIMP-2 are negative regulators of invasion and metastasis of gastriccarcinoma, and are new indices of biological behavior of gastric cancer. TIMP-1 is anindex for predicting prognosis.
