ObjectiveTo investigate the effect of ALDH2gene PolymorPhisms on suscePtibility of stroke,single nucleotide PolymorPhisms(SNP) of ALDH2were investigated in Patients withischemic stroke in Tai’an, Shandong Province.MethodBlood samPles were collected from all subjects including154stroke Patients(cerebral infarction116case,cerebral hemorrhage38cases) diagnosed in88thhosPital and148healthy PeoPle (male68cases, female80cases) as control.The gene PolymorPhismsof ALDH2gene Glu504Lys were detected by laboratory methods and DNA microarraymethod, and were verified by high-throughPut sequencing methods of96-channel caPillaryelectroPhoresis. Information of the the subjects, such as gender, age, birthPlace, smokingand drinking history, medical history including hyPertension, diabetes, coronary heartdisease, the tyPe of carotid vulnerable Plaque Presence and other basic clinicalinformations was recorded to be analyzed using logistic regression analysis of SPSS13.0statistical software.Result1A laboratory method for detecting ALDH2gene Glu504Lys PolymorPhism wasestablished successfully.2The distribution of ALDH2gene Glu504Lys PolymorPhisms was analyzed byHardy-Weinberg genetic equilibrium.3Disribution ratio of A allele of ALDH2genotyPes in the ischemic stroke grouP washigher significantly than that in control grouP.4The distribution of A allele of ALDH2genotyPe in cerebral infarction was increasedsignificantly comParing with that in control grouP(P<0.05) and in cerebralhemorrhage(P<0.05). ALDH2gene mutation and allele A were related to the risk of cerebral infarction.5Univariate logistic regression analysis was emPloyed to analyze the imPortance ofALDH2PolymorPhism. And the imPortance of alcohol consumPtion, hyPertension,diabetes, coronary heart disease and ALDH2PolymorPhism was analyzed by multivariatelogistic regression analysis. It was suggested that smoking, alcohol consumPtion,hyPertension, diabetes, coronary heart disease be indePendent risk factors of stroke, andthe imPact of ALDH2gene PolymorPhism on stroke was not as imPortant as these factors.6In Patients (66cases) suffered from cerebral infarction,we found that75.76%(50/66)had atherosclerotic Plaque examined by carotid ultrasound,among which the vulnerablePlaque was46.97%(30/50). The occurrence of cerebral infarction is associated with theformation of atherosclerotic Plaque, while the Plaque tyPe is mainly vulnerable Plaques.Conclusion1ALDH2gene Glu504Lys PolymorPhism exists in Tai’an area.2ALDH2gene PolymorPhism and the distribution of allele A in individuals withcerebral thrombosis or cerebral embolism were higher significantly than that of healthyPeoPle. HyPertension, diabetes, coronary heart disease,smoking, alcohol consumPtion,agewere associated with the suscePtibility to ischemic stroke,while the imPact of ALDH2gene PolymorPhism on stroke was not so imPortant as the above risk factors.
