The most imPortant cause of stroke,is atherosclerotic artery, coronary heart disease, hyPertension, Previous the second familyhistory of the disease, high blood fat, these factors can lead to the occurrence of cerebralaPoPlexy. After the current ePidemiological investigation of various disPlay, stroke is notalways caused by single genes, living environment and genetic interaction, Promote disease.Prevention of stroke so want better, to exPlore the mechanisms of genetic asPects of veryPromising. The selected grouP is based on the PrinciPle of stroke tyPes, using DNA chiPtechnique, according to the different exPression amount of every gene to select the genes,onthis basis, using real time quantitative PCR method to verify the different exPression genes.By the results we try to demontation the molecular genetic mechanism of different tyPes ofstroke family aggregation,which may become a Powerful basis for the Prevention andtreatment of individual stroke.Methods:ExPeriment from December2012to December2013in Taishan Medical Universityaffiliated hosPital,10Patients with cerebral ischemia in the family (including the firstlevied) over the family cerebral aPoPlexy Patients with75%of the total number of10Patients with cerebral hemorrhage in the family of (including first levied) more than75%of the total number of Patients with cerebral aPoPlexy by the family in addition to select6during the same Period in our kindergarden without a family history of cerebrovascular disease is normal as the control grouP, control grouP and exPerimental grouP with nation,balanced sex ratio, age difference is not more than five years old, and at the same agegrouP. Case grouP and the control grouP a total of26Patients. Using uniformquestionnaire for the selected family and investigation of the Proband and his relatives,record all general clinical data of the object of study, has obtained the consent of all thetested object samPles, using DNA chiP technique, according to the different exPressionamount of every gene to select the genes,on this basis, using real time quantitative PCRmethod to verify the different exPression genes. By the results we try to demontation themolecular genetic mechanism of different tyPes of stroke family aggregation.Results:1. ComPared to cerebral hemorrhage grouP and ischemic grouP, diastolic bloodPressure levels increased significantly (P <0.05); Cerebral ischemia, cerebral hemorrhagegrouP comPared with control grouP in age there was no statistically significant difference(P>0.05), cerebral ischemia, cerebral hemorrhage grouP and control grouP in the historyof history of diabetes, smoking, drinking, systolic Pressure, diastolic blood Pressure,fasting glucose, total cholesterol, low-density liPoProtein cholesterol (hdl-c) weresignificantly increased, and high density liPoProtein decreased significantly, comParedwith the control grouP was statistically significant difference (P <0.05).2. The exPression of gene RXRα was down-regulated in the grouP of Ischemic Stroke,ComPared with that in normal control grouP and in the grouP of hemorrhagic stroke,, andthe significant difference had statistical significance;3. The exPression of gene EVL was uP-regulated in the grouP of Ischemic Stroke,ComPared with that in normal control grouP,and was uP-regulated in the grouP ofhemorrhagic Stroke, ComPared with that in normal control grouP, and the significantdifferences had statistical significance;4. The exPression of gene SYK was down-regulated in the grouP of Ischemic Stroke,ComPared with that in normal control grouP,and the significant difference had statisticalsignificance; but had no significant difference in the grouP of hemorrhagic stroke;5. The exPression of gene lck was down-regulated in the Ischemic Stroke grouP,ComPared with that in normal control grouP,and the significant difference had statisticalsignificance.6. The exPression of gene ID3was uP-regulated in the Ischemic Stroke grouP andHemorrhage Stroke grouP, ComPared with that in normal control grouP, and thesignificant differences had statistical significance. Conclusion:1. The history of history of diabetes, smoking, drinking, blood Pressure, blood fatare a risk factor for Ischemic Stroke and Hemorrhage Stroke; Hemorrhage Stroke, diastolicblood Pressure level is higher than the cerebral ischemia.2. The gene RXRα may had relationshiP with the Pathogenesis of Ischemic Stroke.low exPression may increase its suscePtibility.3. The gene EVL may had relationshiPs with the Pathogenesis of Ischemic Strokeand Hemorrhage Stroke. Over-exPression may increase its suscePtibility to IschemicStroke and Hemorrhage Stroke.4. The gene SYK may had relationshiP with the Pathogenesis of Ischemic Stroke.low exPression may increase its suscePtibility.5. The gene LCK may had relationshiPs with the Pathogenesis of Ischemic Stroke.Low-exPression may increase the suscePtibility to Ischemic Stroke.6. The gene ID3may had relationshiPs with the Pathogenesis of Stroke.UP-exPression may increase the suscePtibility to Ischemic Stroke and Hemorrhage Stroke.
