Objective: To explore the role of Fas and Caspase-8inglioma neoplastic process by examing their expression in gliomatissues.Methods:1. Experienmental group contained90cases ofgliomas specimens confimed by pathology department which were derivedfrom biopsy specimens belonging to patients undergoing craniotomy inNeurosurgery Department of the Affiliated Hospital of LuZhouMedical College during April2012to April2013. There were no anytumors in other parts of body in patients who accepted operation.No radiotherapy or chemotherapy before craniotomy. Control groupcontained20cases of normal brain specimens which were all frompatients undergoing craniotomy because of severe traumatic brain injury;2. The glioma specimens were classified by the2007WHO classification oftumors of the central nervous system after prepared and stained by HE;3.Normal brain specimens were frozen and fixed within12hours after operation;4.examing the expression of Fas and Caspase-8in both groups byimmunohistochemical streptavidin perosidase staining;5.observing andcounting the staining intensity and percentage of positive cells in thenormal brain tissues and glioma tissues.Results:1.Fas was mainlyexpressed in the cell membrane.The positive expression was yellow orbrown yellow in the membrane.There was statistically difference between the expression of Fas in gliomas and in normal brain tissue(Z=-5.948,P=0.000<0.05), and the expression of Fas in normal braintissue was lower than in glioma tissue;2.With the malignance ofgliomas increased, the positive rate of Fas protein increased(rs=0.444,P=0.000<0.05). The statistically differences existed among theexpressions of Fas at all levels of gliomas(χ2=21.038,P=0.000<0.05).The expression of Fas in gliomas of grade Ⅲ(Z=-3.112,P=0.002<0.0083)and grade Ⅳ(Z=-3.586,P=0.000<0.0083)were obviouslyhigher than grade Ⅰ,the differences were statistically significant.There was statistically differences between the expression of Fas inglioma of grade Ⅱand grade Ⅳ(Z=-3.212,P=0.001<0.0083),and theexpression of grade Ⅳ was higher than grade Ⅱ;3.With the peritumoraledema aggravated,the expression degree of Fas proteinincreased(rs=0.538,P=0.000<0.05). The significant differences existedamong the expressions of Fas in gliomas of all levels of peritumoraledema(χ2=29.329,P=0.00<0.05).The expression of Fas in theperitumoral edema of gradeⅡ(Z=-3.240,P=0.001<0.0083) and gradeⅢ(Z=-5.070,P=0.000<0.0083)were higher than grade0,the differences werestatistically significant. There was statistically difference betweenexpression of Fas in the peritumoral edema of grade Ⅰ and gradeⅢ(Z=-2.987,P=0.003<0.0083),the expression of gradeⅠ was lower than gradeⅢ;4.Caspase-8protein was mainly expressed in the cell cytoplasm.The positive expression was yellow or light yellow in the cytoplasm.Therewas statistically difference between the expression of Caspase-8protein in gliomas and in normal brain tissue(Z=-2.101,P=0.036<0.05), and the expression of Caspase-8protein in glioma tissue washigher than in normal brain tissue;5.With the malignance of gliomasincreased, the positive rate and staining intensity of Caspase-8proteindecreased(rs=-0.564,P=0.000<0.05). The statistically differences existedamong the expressions of Caspase-8protein at all levels of gliomas(χ2=35.935,P=0.000<0.05).The expression of Caspase-8protein ingliomas of grade Ⅲ(Z=-3.746,P=0.000<0.0083) and grade Ⅳ(Z=-3.623,P=0.000<0.0083)were obviously lower than grade Ⅰ,thedifferences were statistically significant. The expression ofCaspase-8protein in gliomas of grade Ⅲ(Z=-4.256,P=0.000<0.0083)and grade Ⅳ(Z=-4.276,P=0.000<0.0083)were obviously lowerthan gradeⅡ,the differences were statistically significant;6.With theperitumoral edema aggravated,the expression degree of Caspase-8protein decreased(rs=-0.810,P=0.000<0.05).The significant differencesexisted among the expressions of Caspase-8in gliomas of all levelsof peritumoral edema(χ2=59.814,P=0.000<0.05).There were statisticaldifferences in expression of Caspase-8protein between any twogroups except grade0and gradeⅠ(Z=-1.816,P=0.069>0.0083);7. Thesignificant difference existed in the expressions of Fas and Caspase-8 protein in gliomas(Z=-6.971, P=0.000<0.05) and a negative correlationexisted between the expression of Fas and Caspase-8(rs=-0.662,P=0.000<0.05).Conclusion:1.The expressions of Fas in normal braintissues were lower than in glioma tissues.With the malignance of gliomaand peritumoral edema increased,the expression of Fas increased.Itprompts that abnormal expression of Fas is possibly involved in thedevelopment of glioma and development of peritumoral edema byreduce the apoptosis of glioma cells or promote the proliferationof glioma cells;2.The expression of Caspase-8in glioma tissuewas higher than normal brain tissues. With the degree of peritumoraledema and malignance of glioma increased,the expression ofCaspase-8decreased. It prompts that abnormal expression ofCaspase-8is possibly involved in the proliferation and invasivenessof glioma cells by reduce the apoptosis of glioma cells.In conclusion,there is a close relationship between Fas andCaspase-8.They are possibly involved in pathological grading anddevelopment of peritumoral edema of gliomas possibly by affectingapoptosis of glioma cells.The further research may provide anexperimental basis for a new inhibitory treatment of gliomas.
