Objective: To explore the role of programmed cell death5(PDCD5) and X-linked inhibitor of apoptosis protein (XIAP) inadenomyosis (AM) by immunohistochemistry staining for the clinicaldiagnosis and prevention of experimental data and theoretical basis.Methods:30uterus samples were collected from patients withadenomyosis, then the ectopic endometrium was taken as ectopicendometrium group(Ec group)and the eutopic endometrium which wasfar away from the ectopic endometrium was taken as eutopicendometrium group(Eu group).30uterus samples were collected frompatients with fibroid and the eutopic endometrium which was far awayfrom the fibroid was taken as control group.The apoptosis of endometrialglandular epithelium in these groups was detected by TdT-mediateddUTP Nick-End Labeling(Tunel) assay and the expressions of PDCD5and XIAP were detected by immunohistochemistry assay.Analysis themean optical density(MOD) of PDCD5and XIAP in positive cells byImage Pro6.0. Statistical analysis: data was analyzed with SPSS17.0andthe significant level was defined as P<0.05. Results:1. The apoptosisindex presented a increasing tendency in ectopic, eutopic and controlendometrium. The apoptosis index in Ec group was a little lower thanthat in Eu group(P>0.05) and was obviously lower than that in control group(P <0.05). The apoptosis index in Eu group was obviously lowerthan that in control group(P <0.05).2. The expression of PDCD5in Ecgroup were obviously lower than that in Eu group(P <0.05) and controlgroup(P <0.05). The expression of PDCD5in Eu group was obviouslylower than that in control group(P <0.05) and a little higher than that inEc group(P>0.05).3.The expression of XIAP in Ec group were obviouslyhigher than that in Eu group(P <0.05) and control group(P <0.05). Theexpression of XIAP in Eu group was obviously higher than that in controlgroup(P <0.05) and a little lower than that in Ec group(P>0.05).4.Therewas a negative correlation between PDCD5and XIAP both in Ec group(r=-0.415, P=0.023<0.05)and Eu group(r=-0.382, P=0.037<0.05).There was no correlation between PDCD5and XIAP in control group(r=-0.126, P=0.506>0.05). Conclusion:1.Compared with the controlgroup, the apoptotic indexes of Ec group and Eu group were significantlyreduced, which indicated that the abnormal endometrial cell apoptosismaybe indeedly exist in the formation of AM. This anomal endometrialcell apoptosis maybe an important factor in the formation of a direct orindirect participation AM.2.Compared with control group, theexpressions of PDCD5and XIAP in Ec group and Eu group weresignificantly different. The significant down-regulation of PDCD5andsignificant up-regulation of XIAP suggested that the abnormalexpressions of PDCD5and XIAP maybe the important factor of resulting in the development of anomal endometrial cell apoptosis inadenomyosis.3. There was a negative correlation between PDCD5andXIAP both in Ec group and Eu group, which suggested there may be anegative feedback or antagonist relationship between PDCD5and XIAP.Besides, the expression of PDCD5was lowest in Ec group and theexpression of XIAP was highest in Ec group, which further suggested thatthere may be a significant negative feedback or antagonist relationshipbetween PDCD5and XIAP in Ec group so as to break out the balance ofapoptosis, and they may jointly promote the development ofadenomyosis....
