Abstract:objective:Angiopoietin-like protein3(ANGPTL-3) can regulate angiogenesis. Our purpose of this experiment is to analyze the correlation of the concentration of ANGPTL-3in the serum and vitreous, and provide a new method to the prevention and treatment for proliferative diabetic retinopathy (PDR), by quantitative detecting the expression of ANGPTL-3in the serum and vitreous of the PDR, and analyzing the differences of ANGPTL-3concentration in serum and vitreous, Methods:Seleced PDR patients28cases who required standard three channel vitrectomy, including10males,18females, between28and68years old, average age of50.75years;5cases retinal detachment with vitreous hemorrhage associated,14cases vitreous hemorrhage without retinal detachment,9cases retinal detachment without vitreous hemorrhage.The control group enrolled14cases who had idiopathic macular hole or rhegmatogenous retinal detachment and so on, and required standard three channel vitrectomy, including4male and10female, between16and71years, average age of51years;5cases macular hole with retinal detachment,6cases macular hole without retinal detachment,3cases with rhegmatogenous retinal detachment. Recording the patient’s basic information on admission, all patients were required to measure blood pressure, blood glucose (fasting blood glucos,random blood glucose), blood lipid. The PDR patients needed to bemeasuring glycated haemoglobin (HbA1c), blood glucose two hours aftermeals. In morning pumped3ml elbow venous blood, centrifuged at4℃and3000rpm for10min, separated the supernatant with aliquot thesample, stored at-80℃ultra low temperature freezer immediately to bemeasured. The fasting blood glucose of the PDR patients should lowerthan8.3mmol/L at the operating day. In the three channel vitreousstandard resection while closed perfusion acquisition central vitreoushumor about0.6-1.0ml, centrifuged at4℃and15000rpm for5min,separated the supernatant immediately, stored at-80℃ultra lowtemperature freezer for prepared. Vitreous and blood samples ofcentrifugal speed were not the same, in order to better filter sediment, soas to the test results not interfered. Using enzyme linked immunosorbentto detect the concentration values of ANGPTL-3in the serum andvitreous in two groups patients. Using SPSS17.0statistical softwaresystem to statistical analysis all the data, all the normal distribution datawas expressed as average±standard deviation (ˉx±s), using T test of twoindependent samples, and the skewed distribution data using rank sumtest (Z test), and Spearman correlation analysis was used for thecorrelation between the two samples. P<0.05indicated as significantdifference. Results: The difference of the age and gender beteen the PDR group and the control group was not statistically significant(P=0.679>0.05). Compared with the control group, the difference of thevitreous ANGPTL-3in the PDR groups was not statistically significant(P=0.178>0.05). Compared with the control group, the difference of theserum ANGPTL-3in the PDR group was statistically significant(P=0.045<0.05). There was no correlation between the ANGPTL-3in theserum and vitreous in the PDR group (r=-0.1134, P=0.5656). There wasno correlation between the ANGPTL-3in the serum and vitreous in thecontrol group (r=-0.5075, P=0.064). Conclusion: ANGPTL-3may relateto the occurrence of diabetes; ANGPTL-3may paly little role in thedevelopment of the PDR.
