Effect Of Shuyu Capsule On5-HTR1A-PKA-CREB Signal Pathway In Hippocampus Of Rats With Depressive Disorder

Objective:To investigate the5-HTR1A mediated PKA-CREB signaling pathwayinvolved in pathogenesis of depressive disorder, and to study the signal transductionpathway is a target of Shu Yu capsule.Methods: The rats were divided randomly into normal group, model group, Shuyugroup, fluoxetine group, antagonist group. Using the chronic unpredictable mild stress(CUMS) to prepare model and behavior tests to evaluate rats. The expression levels of5-HTR1A, PKA, p-PKA, CREB, and p-CREB were detected respectivly with Westernblot.Results:1、Compared with the normal group, body weight, open field test score,sucrose preference values of rats in the model group decreased significantly (P <0.05).Those of rats in Shuyu group, fluoxetine group and antagonist group rats increasedobviously compared with the model group (P <0.05). There was no significantdifference between rats in Shuyu group, fluoxetine group and the antagonist group.2、The Western blot results showed that hippocampal5HTR1A protein levels ofmodel rats increased significantly (P <0.01) compared with those of rats in normalgroup. The5HTR1A protein levels of rats in Shuyu group, fluoxetine group and theantagonist group decreased significantly (P <0.01) compared with those of rats in themodel group. The phophrylation levels of hippocampal PKA and CREB in rats ofmodel group significantly decreased (P<0.01) compared with those of rats in thenormal group. While those of rats in the Shuyu group, fluoxetine group and theantagonist group ramarkably increased (P<0.01) compared with the model group rats. Conclusion: Shuyu capsule can effectivly correct behaviour changes of derpressivedisorder rats. The5-HTR1A mediated PKA-CREB signaling pathways in hippocampusis one of the targets of Shuyu capsule in depressive disorder treatment.
