Purpose: Based on the mice of CMUS depression model and C-FOS protein expressing of hippocampus area,the study namely discusses the anti-depression mechanism of action of Shuyu Granule to mice of CMUS depression model and therefore cure clinical depression.This thesis studies the ethological effects on the mice of CMUS depression model of Shuyu Granule by experiments,proves its anti-depression results and explicits the dose-effect relationship.The thesis also studies whether the anti-depression effects of Shuyu Granule which bears “relieve the mental stress”as its principle could improve the C-FOS protein expressing in hippocampus area from the theoretical level,thus offering the theoretical basis.Methods:The study adopts multi-factor chronical and unpredictable stress stimulation includes:abrosia,water-break,damp bedding,cage shocking+ noise stimulation,tilted cage,ice water swimming and tail clipping,etc.In these way can we build chronical and unpredictable stress stimulation depressed mice.Then we offer them Shuyu Granule by different dose.The study divided Kunming mice into Blank group,Model group,Positive drug fluoxetine group and Shuyu Granule group of high,mild and low dose and observed their behavioral changes by free activities,tail suspension test,compelling swimming test and sweet water favored test.The study also adopts immunohistochemical method to observe the C-FOS protein expressing in hippocampus area and therefore further discusses the behavioral changes of chronical and unpredictable stress stimulation mice and anti-depression action mechanism of Shuyu Granule.Results:1.The body mass of experimental animals in comparison group and medical cure group all grew.However,the body mass of CUMS depression mice grew more slowly than its of Blank group;Compared to CUMS depression mice,the food ration of other medical cure groups had a obvious grow.Food ration:Blank group had a steady raise;Model group was a little bit low compared to Blank group;Medical cure groups enjoyed an obvious grow.2.Statistics of free activities showed that Blank group was in stable condition,CUMS model mice had less activities.And after the medical cure,the activities of mild and high dose grew compared to model group.3.In tail suspension test,the mice of CUMS model group held still for more time than those in Blank group.And compared to model group,mice of medical cure groups stayed still for a relatively short time.4.In compelling swimming test,the motionless time of CUMS model group obviously grew compared to blank group.Compared to model group,the motionless time shrank in medical cure groups.5.In sweet water favored test,the sweet water consuming volume of CUMS model group decreased compared to blank group.Compared to model group,consuming volume of the medical cure groups grew.6.In blank group,the C-FOS positive cell expressed a little.In model group,the C-FOS positive cell expressed less than blank group.In medical cure groups,the C-FOS positive cell expressed more than model groups and less than blank group.Conclusions:1.Shuyu Granule could obviously improves the behavioral changes of unpredictable stimulation depression mice and presents certain anti-depression effects.2.Shuyu Granule could fight against the decrease of C-FOS expressing in area of the mice and relieves the neuronic damages.Chinese Herbal Compound Shuyu Granule possesses certain anti-depression effects,its therapeutic mechanism might has a relationship with gene expressing of central neurons C-FOS.
