Objective:Exploring the effect of Huanglian Jiedu Decoction on middlecerebral artery occlusion (MCAO) in rats with acute lung injury ((ALI) experiment andstudy of its mechanism.Methods: Healthy male Wistar rats were randomly divided into blank group, shamoperationgroup, model group, Huanglian Jiedu Decoction high dose group, low dosegroup,Each group was divided into1D,4D,7d group with three subgroups, a total of15groups.Using MCAO model, to observe the effects of different dose in different time insolution on bronchoalveolar lavage fluid (BALF) changes, inflammatory cell counts inlung tissue of pulmonary pathological changes and ultrastructural morphological changes.Results:(1)In MCAO model group, BALF (bronchoalveolar lavage fluid) in total cells,monocytes, lymphocytes, neutrophils, eosinophils were significantly higher than that ofthe control group and the healthy control group, there was significant difference (P<0.01);BALF Huanglian Jiedu Decoction high dose group and low dose group of total cells,monocytes cells, lymphocytes, neutrophils, eosinophils were significantly lower thanthose in the model group, but higher than that of the control group and the healthy controlgroup, there was significant difference (P<0.01); and Huanglian Jiedu Decoction high dose group and low dose group, the total number of cells in BALF monocyte, lymphocyte,neutrophil, eosinophils array with no significant difference(P>0.05).(2) In lung tissue of rats with mild infiltration of inflammatory cells, capillaries in asmall amount of red blood cells, alveolar septal thickening in the lumen of mild, almostno epithelial cell shedding; sham operation group of inflammatory cells in lung tissueswith a little exudate within alveolar walls, a large number of inflammatory cells,eosinophil, neutrophil, lymphocyte and macrophages, capillary a few red blood cells,alveolar septum slightly widened lumen visible, suspicious and shedding of epithelialcells; normal control group had no obvious pathological changes; Huanglian JieduDecoction high dose group and pathological change is lighter: inflammatory cells have alittle exudation, capillary in a few red blood cells, alveolar space slightly a widenedlumen visible, suspicious and shedding of epithelial cells; change of Huanglian JieduDecoction low dose group of lung tissue is light: inflammatory cells have a littleexudation, capillary in a few red blood cells, alveolar septum slightly widened lumenvisible, suspicious of exfoliated epithelial cells. Model group and blank control group andsham operation group were statistically significant (P <0.05), Huanglian Jiedu Decoctionhigh dose group and low dose group and model group were statistically significant (P <0.05), and high dose group than in low dose group was significantly more ease.(3)Normal control group: alveolar type II epithelial cell lamellar body compact, normalstructure; alveolar cavity microvilli protrusions arranged in neat rows; mitochondrialcristae; structural integrity. Sham operation group: alveolar type II epithelial cell lamellarbody part of emptying; alveolar cavity slightly exudate, mitochondria swelling andrelatively obvious. The alveolar surface microvilli decreased than that of the normalcontrol group, but were arranged. Model group: the structure of alveolar type II epithelialcells, cell structure is not clear, the lamellar body obvious vacuolization; mitochondria swelling, cristae broken or disappeared; the cell surface microvilli decreased and evendisappeared. Huanglian Jiedu Decoction high dose group of alveolar type II epithelial celllamellar body part of emptying shaped mitochondria swelling is not obvious. Alveolarcavity with a small amount of exudate, surface microvilli than in the normal controlgroup reduced slightly, but arranged in neat rows of Huanglian Jiedu Decoction low dosegroup of alveolar type II epithelial cell lamellar body part of emptying shape; a largenumber of alveolar cavity exudate, mitochondrial swelling. Compared with the normalcontrol group, surface microvilli decrease slightly, arranged irregularly..Conclusion(1)Huanglian Jiedu Decoction intervention can reduce inflammatory cell infiltration ofacute lung injury in MCAO rats, the MCAO rats lung injury。(2)Huanglian Jiedu Decoction intervention can reduce inflammatory cells in lung tissueof acute lung injury in MCAO rats infiltration, improve capillary hemorrhage, congestion,reduce the lumen almost no epithelial cell shedding, on the MCAO of rat lung injury(3)Huanglian Jiedu Decoction intervention can protect acute lung injury of MCAO ratslung tissue subcellular structure...
