Study On The Treatment Of Acute Rat Model With Hemorrhoids By.Self-made “Huanglian Jiedu Decoction”

Objective:By observing the effect of self-made Huanglian Jiedu Decoction on the general situation?perianal ulcer?Perianal pathology?inducible nitric oxide synthase(i NOS)?Vascular endothelial growth factor(VEGF)?Prostaglandin E2(PGE2)of acute rats model with hemorrhoids,to explore the efficacy and action mechanism of self-made Huanglian Jiedu Decoction,which can provide non-surgical selection for the treatment of acute hemorrhoids and clinical basis for the use of Huanglian Jiedu Decoction.Methods:Prepared 44 SD rats,and treated with perianal injection of glacial acetic acid to replicate the acute model of hemorrhoids,and then randomly selected 4 from these models by observing the Perianal pathology to confirm the building success or not.The remaining 40 rats were randomly divided into experimental H1 group(high dose),experimental H2 group(low dose),experimental control H3 group(Mayinglong hemorrhoids ointment)and model control H4 group(distilled water),10 per group.The H1,H2 group were given respectively the Huanglian Jiedu Decoction high doseand low doseby fumigation method;The experimental control H3 group was given Mayinglong hemorrhoids ointmentby smear method;The model control H4 group was given sterile distilled water by fumigation method,the above groups were given twice a day interval of 4 hours,and the treatment period was 7 days.The general situation was observed during the period of drug delivery,the perianal ulcer area and swelling degree were recorded on the second day,the fourth day,the seventh day.Took perianal ulcer tissue for pathological observation and immunohistochemical staining was used to analyze the positive rate of i NOS?VEGF?PGE2 after the end of the administration.Results:1)The rats in all groups were in good condition.2)Localized inflammation score,perianal local specimen pathological examination were consistent with hemorrhoid acute phase performance.3)The local inflammation score and the ulcer area score were no significant difference between the experimental H1 group,the experimental H2 group,the experimental control H3 group and the model control H4 group before medication(p>0.05),which was comparable.4)At the end of the administration,the following conclusions were drawn for each group: High concentration of decoction to promote the healing of perianal ulcer was significantly better than the role of Mayinglong hemorrhoids ointment(p<0.05);Low concentration of decoction to promote the healing of perianal ulcer was significantly better than the role of Mayinglong hemorrhoids ointment(p<0.05);The effect of low-concentration decoction and high-concentration decoction on the healing of peripartumal local ulcer was statistically insignificant(p>0.05).5)The inhibitory effect of high concentration decoction on iNOS,VEGF and PGE2 expression was better than that of Mayinglong hemorrhoids ointment(P<0.05);The inhibitory effect of low concentration decoction on i NOS,VEGF and PGE2 expression was better than that of Mayinglong hemorrhoids ointment(P<0.05);The inhibitory effect of low concentration decoction and high concentration decoction on iNOS,VEGF and PGE2 expression was statistically no significant difference(p>0.05).Conclusion:1)The Self-made Huanglian Jiedu Decoction can improve the symptoms of edema and ulcer in the rat model of the similar animal model,and in this experiment,the effect of the high and low dose decoction on the rat model of the similar symptoms was not significant;2)The self-made Huanglian Jiedu Decoction can improve the pathological symptoms of rat model of hemorrhoids,and can inhibit the inflammatory reaction and promote the healing of ulcer;3)The self-made Huanglian Jiedu Decoction in the treatment of acute stage of animal model of hemorrhoids may be related to iNOS,VEGF and PGE2 factors;4)VEGF can promote the growth of blood vessels,and speculated that VEGF has some correlation with the formation of hemorrhoids;5)This experiment confirmed that the self-made Huanglian Jiedu Decoction with fumigation on rat model of similar hemorrhoids curative effect,at the same time provide a non operative treatment for the clinical treatment of hemorrhoids.
