OBJECTIVE A comparative study of antidiabetic effect about Compound fenugreek seed extract in two models of insulin resistance with glucose and lipid metabolism disturbance.To explore the relationship between CFSE and antidiabetic effect in the model of insulin resistance with glucose and lipid metabolism disturbance by dexamethasone. These exploration will pray the way for the in clinical application of CFSE.METHODS The way of rats insulin resistance with glucose and lipid metabolism disturbance is based on" security of the State Food and Drug Administration [2012] No.107" file,compared with the normal. An oral glucose-tolerance test is needed before the end of the experiment.The FBG,OGTT/TC,TG,FINS,HbAlc,HDL and LDL were carried out after fasting.The expressions of IRS1,GLUT4and PPARy were examined using Western Blot method.RESULTS Both methods can be set up the insulin resistance with glucose and lipid metabolism disturbance. Specific performance in the 0.5hours blood glucose, there is a significant difference to the normal control group(P>0.05). These two models have no significant difference between model control group and normal control group in FBG,2hours blood glucose and AUC of glucose(P<0.05).The TG and TC value of MC group significantly higher than the NC group(P<0.01), the HOMO-IR has no significant difference between MC group and NC group(P>0.05), described above, these two models of insulin resistance with glucose and lipid metabolism disturbance is set up successfully. For the model of insulin resistance with glucose and lipid metabolism disturbance by dexamethasone,CFSE has a significant difference in the impaired glucose tolerance(P<0.05) and has no significant difference in TC,TG,FINS,HOMO-IR,HbAlc,HDL,LDL value(P>0.05) compared with MC group. For the model of insulin resistance with glucose and lipid metabolism disturbance by alloxan,CFSE group has no significant difference in the impaired glucose tolerance and TC,TG value(P>0.05). Except low-dose CFSE group has a statistical significance in HDL value to the rats of insulin resistance with glucose and lipid metabolism disturbance by alloxan(P<0.05),CFSE has no statistical significance in FINS,HOMO-IR,HbAlc,HDL and LDL value to both of models. The expressions of IRS1protein on each1concentration CFSE group has no significance in the muscle tissues(P>0.05),but the expressions of GLUT4protein in the muscle and fat tissues and the expressions of PPARy protein in the fat tissues is significantly higher than MC group(P<0.05).CONCLUSION The two models of insulin resistance with glucose and lipid metabolism disturbance in rats has been established successfully. CFSE has an antidiabetic and improving insulin resistance effect in the model of insulin resistance with glucose and lipid metabolism disturbance by dexamethasone.Increasing the expressions of PPARy protein may be one of the major mechanism,which can inhibit the cytokine release,activate the signal transduction on PI3K and increase the expressions of GLUT4protein.However, the end result will be an antidiabetic effect and improving insulin resistance.
