Objective:Bone growth factors can regulate the metabolism of bone growth, toimprove clinical therapeutic effects of dental implants it will be applied tothe clinical. This experiment aims to establish bone defect model withimplant implanted.Compare the effect of PDGF, IGF-1and combinedon the bone repair, and to explore the effect of bone growth factorsPDGF and IGF-1promoting bone repair around the pure titaniumimplants.Methods:(1) Chosing male or female rabbit24and average weight2.3kg.(2) Anesthetizing rabbit with chloral hydrate,clean the skin,disinfection, spreading drapes,separating skin incision step by step, fully exposing two iliums, manufacturing rabbit iliac defects with cylindricalstainless steel needle,implanting pure titanium implants,producing alveolar bone implanted model.(3) Then they were randomly divided into4groups,respectively IGF-1,PDGF-BB,IGF-1+PDGF-BB groups and negative physiological saline ascontrol groups. (4) And the animal were killed after three days,seven days and tendays,and then take out the implants and the soft tissue arroud theimplants,and HE staining.Results:1、Gross observation:3days after operation,mental state of all the rabbitbecomes poor,diet activity decreases,limb slightly swollen,and walk withdifficulty. At the fourth day,all the rabbits return to normal activities, dietand mental both in good state.No implant has been discharged or takenoff, implant and the surrounding tissue connect closely.2、Histology observation:In IGF-1group granulation tissue form obviously, giant cells exist abundantly,a small amount of calcium saltdeposits and kind of bone tissue appear. In PDGF-BB group, granulation tissue is also visible, a little calcium salt deposits and thekind of bone tissue forms.In IGF-1+PDGF-BB combination group, granulation tissue has become a fibrous connective tissue, boneand calcium salt deposits formed. In physiological saline group noobvious granulation tissue formation, local tissue necrosis, inflammatory cells infiltration.Conclusion:1. Applying single factor, in IGF group it play an important role in boneconnection surrounding the implant. PDGF-BB has an effect on boneconnection surrounding the implant only in short-term. 2. Applying the two factors, PDGF-BB+IGF play the biggst role inosseous junction around implant, calcium salt deposit is not obvious,thickness of damage is small,damage area is small. The synergy of PDGFand IGF1promote bone around implant integration function, and theycan accelerate bone formation.
