| Monocyte Chemotactic Protein3(MCP-3,also known as CCL7) is one of theseparated cloning CC chemokines and family,It is the strongly activation andchemokine of mononuclear cells and macrophages, and participate in the developmentof a variety of inflammatory disease and cell pathological state.MCP-3mainly causesmononuclear cells and macrophages gathering to inflammation place continuedly andactivates monocytes and macrophages playing a role of engulf pathogens and antigenpresenting. MCP-3also has chemotactic activity of lymphocytes, natural killer (NK)cells and dendritic cells (DC) and basophilic cells,inducing the inflammatory cells togather to the inflammation area,causing a series of inflammatory reaction.In view ofMCP-3can activate a variety of cells involved in inflammation,therefore, currently,MCP-3become a hotspot research in the field of inflammation, tumor, etc.Recently, foreign medical research from various disciplines in the field ofdifferent inflammatory disease found MCP-3associated with the development ofvarieties of chronic inflammatory diseases,the pathological state of abnormalexpression of MCP-3, can lead to allergic disease,atherosclerosis,enteritis,cervicalcancer,myocarditis ect.many other clinical diseases.Related research in the field oforal diseases confirmed that MCP-3periarthritis with the development of apex andchronic diseases such as periodontitis, oral cancer. Pulpitis, as one of the commoninflammatory diseases of the oral cavity, often secondary to bacterial infection causedby tooth decay or retrograde periodontal infection, the inflammation development willinevitably accompanied by the accumulation of inflammatory cells, according to theproperties of the MCP-3chemokines,disorder diseases will also be inevitablyaccompanied by the emergence of the MCP-3and trigger relevant changes ofinflammatory cells in the process of pulpitis.but,as so far, there are no experimentalproof of MCP-3existoing in inflammatory dental pulp cells and its related effect,so this experiment use immunohistochemical method to detect the expression ofMCP-3in the normal dental pulp, acute pulpitis and chronic pulpitis pulp tissue,further study the mechanism of the chemotactic factor MCP-3in the development ofpulpitis, to reveal the role of MCP-3in the development of pilpitis disease, andimprove the chemotactic factor related functions in the course of oral disease.Theresults show that MCP-3has no expression in normal dental pulp tissue, but hasdifferent degree of expression in the inflammatory tissue.Result as follows:1.The group of healthy pulp:there were fibroblasts, collagen fibers,capillary,tissue cells and a few macrophages but no positive expression oforange-brown positive cells stained by MCP-3Under microscopes.2.The group of acute pulpitis:Microscopically observed large Numbers ofneutrophils swam out blood vessels and chemotactic migration to the inflammatoryarea,MCP-3positive cells mainly distributed in neutrophils, monocytes andmacrophages,the average grey value of Positive cell was20.the quantity and degree ofpositive staining cells were disparity,positive expression were mainly distributed inthe cytoplasm and cell membrane and formed tan particles.3.The group of chronic pulpitis:Microscopically observed vascular endothelialcells, macrophages, neutrophils, fibroblasts and various degree positive expression ofMCP-3on the nascent capillaries,the average grey value of Positive cell was25,therewas no significant difference comparing with acute pulpitis organizations(P>0.05),positive expression were mainly distributed in the cytoplasm and cellmembrane and formed tan particles.Above all, MCP-3expression in pulpitis pulp tissues together with other CCchemokines and family play a major role in the inflammatory tissue healing. |
