| Objective: The article makes a study of the Characteristics and the spatial relation between the neuropeptide and antigen-presenting cells in various stages of pulpitis. Namely, through the observation of the Characteristics and the spatial relations of the neuropeptideY of dental pulp, the dendritic cells and the vascular endothelial growth factor in an inflammatory process of rat dental pulp, we can make an approach to the effect of these factors(NPY,DC,VEGF) which will be shown in the inflammatory process of the dental pulp, and make a deeper understanding of the pathology of dental pulp ache.Method: Establishing an experimental dental pulp model of big rats, using the method of immunity set turn and the technique of the picture analysis to examine 40 big rats'molars totally which dental pulp is normal or caught the pulpitis. The molars were divided into two groups: the normal ones and the pulpitis ones. Normal ones are used to set a matched control, and the pulpitis ones is divided into the 3 sets according to time, they are the 1d set, the 3d set, and the 5 d set. Using three polyclonal antibodys(NPY, TY-3H, and VEGF) separately mark the NPY,the DC and the vas in the dental pulp, and make the qualitative analysis and quantitative analysis with the nerve fiber containing NPY, the dendritic cells and the vas.Results:1.The normal groups and the pulpitis groups all show the existing of the NPY, the DC, and the VEGF and encircle the vas.2.The pulpitis groups show more neuropeptideY positive fibers expression than the normal ones, and there are significant differences in comparing with each different set in pulpitis group. (P<0.05)3.In the process of pulpitis, the quantity of the dendritic cells increased according to the development of the inflammation. There are significant differences in comparing with each different set in pulpitis group. (P<0.05) 4. All groups show the expression of the VEGF. The number of positive vessel increased and these vessels become thicker, and the lumen was enlarged according to the development of the inflammation. There are significant differences in comparing with each different set in pulpitis group. (P<0.05)5.Double-immunohistochemical staining shows two different results. The normal group shows no contact between the NPY and the DC, but in the pulpitis groups, The NPY and the DC co-increased in the perforation of pulp chamber region, and assembled under the odontoblasts and around the vas. We can observe that the NPY contacts the DC. The NPY will approach the vas step by step according to the inflammation process.Conclusions:1.Common dental pulp exists the neuropeptide Y and its positive nerve fiber will be increased and enlarged obviously according to the inflammation process and will approach the vas step by step.2. In response to carious stimulus of the pulpitis, the neuropeptide Y and the Dendritic cells will co-increase.3.In the dental pulp, dendritic cells'quantity will be changed according to the inflammation process, when the inflammation grows hard, dendritic cells and positive vases will increase. |
