Objective:To explore the effects of Buyang Huanwu Decoction on neural stem cells after hypoxia injury via the expression of Wntl and Wnt3a, and the possible mechanism of nerve regeneration of ischemic cerebral vascular disease after the effect of Buyang Huanwu Decoction.Methods:1.Obtaining the experimental cell after the establishment of Neural stem cells in vitro culture system from embryonic rat wild mice, and the identification of their proliferation.2.The experimental cells were divided into hypoxia injury+serum of Buyang Huanwu Decoction group (A group), hypoxia injury+serum of control group (group B), normal+serum of Buyang Huanwu Decoction group (C group), normal+serum of control group (D group).3.Through the expression of NF200and GFAP on A group, B group, C group, D group by immunofluorescence detection, studing the effects of Buyang Huanwu Decoction on neural stem cells after hypoxia injury.4.Through the expression of NF200、GFAP、WntlandWnt3a on A group, B group, C group and D group by semi quantitative qRT-PCR detection, further studing on the effects and the possible mechanism of Buyang Huanwu Decoction on neural stem cells after hypoxia injury.Results:1.There are expression of NF200and GFAP on different group by immunofluorescence detection.2.The compared result of the expression of NF200and GFAP in different group by semi quantitative qRT-PCR method is that compared with B,NF200in A were increased obvious (P<0.01);compared with D,NF200in C were increased (P<0.05);C compared with D (P<0.05); compared with B,GFAP in A were increased (P<0.05);compared with D,GFAP in C were increased (P<0.05); Compared with group A, the ratio in the other3groups of NF200and GFAP mRNA relative amount of were decreased (P<0.05); and C group than that in group D, the ratio of NF200and GFAP mRNA was no significant (P>0.05). 3.The compared result of the expression of Wnt1and Wnt3a in different group by semi quantitative qRT-PCR method is that compared with B,Wntland Wnt3a in A were increased obvious (P<0.05);compared with D, Wntland Wnt3a in C were increased (P<0.05); compared with D,Wntland Wnt3a in B were increased obvious (P<0.05)(P<0.05)Conclusion:1.Buyang Huanwu Decoction can promote the differentiation of stem cells after hypoxia injury.2.Buyang Huanwu Decoction can promote the neurons tend differentiation of stem cells after hypoxia injury.3.Wntl and Wnt3a may be involved in the process of Buyang Huanwu Decoction to promote differentiation of stem cells in ischemia.
