Purpose:To observe the influence of Buyang Huanwu Decoction (BYHWD) on the contentof MDA, the activity of SOD and the ultra structure of optic nerves after theoptic nerves of rats have been experimentally injured, it further discussesthe repair effect and functional system of BYHWD on optic nerves injury.Methods:1. Select40normal SD rats, and at random divide5of them into the normalcontrol group, another5into the sham operation group, another15into themodel control group, and the rest15into the treatment group. According todifferent observing time after the optic nerves being injured by clipping,the model control group and treatment group can be divided into3subgroupswith5rats each. These subgroups are5days,14days, and28days.2. Build up animal models of rats with clipped injury on optic nervesin model control group and treatment group, fill BYHWD to the rats in thetreatment group, and fill equal quantity of normal saline to the model controland sham operation group.3. Kill the rats respectively on the5thday, the14thday, and the28thday. Extract optic nerves and measure the content of MDA and the activity ofSOD separately in optic nerves by the methods of thiobarturic acid and xanthineoxidase, and then observe the change of ultra structure of optic nerves throughtransmission electron.Result:1. The content of MDA in the normal control group is2.80±0.08. Comparedwith the normal control group, the content of MDA in the treatment group experienced no obvious increase (P>0.05); the content of MDA in the modelcontrol group increased significantly (P<0.05). Compared with the treatmentgroup, the content of MDA in optic nerves in the model control group hasobviously increased on the5thday, the14thday, and the28thday after operation(P<0.05).2. Compared with the normal control group, the activity of SOD in opticnerves of the treatment group and the model control group both decreasedsignificantly (P<0.05). Compared with the model control group, the activityof SOD in optic nerves of the treatment group is all higher than that in themodel control group on the5thday, the14thday, and the28thday after operation,which reveals obvious statistical distinction (P<0.05).3. Compare the normal control group with the sham operation group, noobvious distinction on the content of MDA and the activity of SOD in opticnerves.4. The myelin sheath structure of the sham operation group is basicallynormal, without pathological changes. The ultra structures of optical nervesin the model control group and the treatment group both changed, for example,vacuoles of mitochondrions degeneration, axon disarrangement, separation ofunconsolidated lamellar, and so forth. But compared with the model controlgroup at the same point of time, the treatment group changed relativelyslightly. The number of myelinated nerve fibers reduced more lightly, thearrangement was more orderly, and the thickness of myelin sheath was more even.Conclusion:BYHWD has some protection effect on experimental optic nerves injury ofrats.
