Objective:To study the protective effect and possible mechanisms of effective fraction from Buyang Huanwu Decoction (EFBHD) against global cerebral ischemia reperfuson injury in rats.Methods:The acute cerebral anoxia mice model induced by decapitation was used to evaluate the anti-hypoxic. The duration of electrocardiograph (ECG) after clamping trachea was observed in mice.Global cerebral ischemia reperfuson injury was caused by four vessel occlusion (bilateral vertebral arteries and bilateral carotid arteries,4-VO) in rats. The electroencephaloographic (EEG) changes were recorded. The content of malondialdehyde(MDA), the activity of lactate dehydrogenase (LDH) and Glutathione peroxidase(GSH-PX) in plasma,and the water content of brain were measured. The cellular morphologic changes were observed by HE staining method. Neuronal cell apoptosis of cerebral tissues was detected with TUNEL method. Thrombus model of artery-veins cross in rats was used. The wet weight and dry weight of thrombus in vitro were measured. Platelet aggregation was studied by turbidimetry.Results:EFBHD (250、125、62.5mg/kg) could significantly prolong mice gasping time after decapitation. EFBHD (250、125mg/kg) markedly prolonged the ECG time after lamping trachea on the cerebral anoxia model induced by clamping trachea in mice. Compared with the model group, EFBHD (200mg/kg) could improve the EEG change of rats by4-VO.EFBHD (200mg/kg) could markedly decrease the MDA content, and EFBHD (200、100、50mg/kg) decrease the LDH activity while increase the GSH-PX activity in plasma,too.EFBHD (200、100mg/kg) significantly reduced the water content of brain induced by global brain ischemia-reperfusion injury in rats. Histopathology examination showed that sham group had no infarction areas and evident pathological change, and in model group, the number of neurons at ischemic area decreased, nerve cell damage by4-VO in rats. And EFBHD could inhibit the apoptosis of nerve cell of cerebral tissue by4-VO in rats. EFBHD could decrease the wet weight and dry weight of thrombosis on the A-V thxombosis by-pass model. EFBHD (200mg/kg) has significant effect on inhibition of platelet aggregation.Conclusion:These results indicate that Effective fraction from Buyang Huanwu Decoction has protective effects against cerebral ischemia reperfuson injury in rats, which might be associated with its antioxidant properties, anti-apoptosis effects, antiplatelet effects and anti-thrombos.
