| Objective:1. Through the research on ancient and modern literatures, to summarize traditional Chinese medical treatment rule and provide an theoretical reasons to treat chloasma about syndrome differentiation.2. Research curative mechanism about modified grease of Shengyu(MGS) for the treatment of chloasma based on the experimental animal model.Methods:1. Retrieve the document of the syndrome differentiation and treatment about chloasma published from1989to2013, analyze dialectical elements: location of disease, nature of disease, cause, therapeutic principle, distribution features of the commonly used formula and so on, summarize traditional Chinese medical treatment rule.2. Using progesterone attack mouse to make chloasma model.3. Using immunohistochemistry (IHC) technology, hydroxylamine method and TAB method to detect distribution and expression about melanin cells, T-SOD activity and MDA content in the skin and liver on chloasma model.4. Using epinephrine combined cold stimulation to mouse to make blood stasis model.5. Using MGS to treat the model of blood stasis syndrome, observe changes of hemorheology before and after treatment.Results:1. Chloasma’s clinical syndrome type is a total of28types, cause of disease:six evils, seven emotions, diet partial addicted, excessive work and others; location of disease:kidney, spleen, lung, heart, bravery, stomach, channels and cellular palace; Nature of disease can be divided into13factors, and the top five factors:qi stagnation, Yin deficiency, blood stasis, blood deficiency and qi deficiency; Therapeutic principle can be divided into26elements; Formula:relief liver for smooth Qi with xiaoyaowan, nourishing liver and kidney with liuweidihuangwan, promoting blood circulation and removing blood stasis with taohongsiwutang, invigorating spleen and supplementing qi with buzhongyitang.2. Liver SOD vitality:there are differences between the blank group and the model group (P<0.05); compared with model group, there are differences between low dose groups and middle dose groups (P<0.05).3. Skin SOD vigor:the low, medium and high dose groups all become higher; there are differences between the middle dose groups and the blank groups (P<0.05); there are differences between middle dose groups and model groups (P<0.01).4. The content of liver MDA:the highest is model group, while the lowest is the low dose group. There was no statistical significance between the groups.5. The content of skin MDA:there is significant difference between the blank group and the middle dose group (P<0.01) and little difference compared with model group. Compared with model group, low dose group has little group (P<0.05) while middle group and high dose group have significant difference (P<0.01).6. High, medium and low dose group compared with the blank group distribution to reduce melanin cells, with a significantly different dose group.7. blood stasis syndrome in rats blood rheology, compared with model control group, high dose group of whole blood viscosity, whole blood high shear reductive viscosity, plasma viscosity, whole blood high shear reductive viscosity, whole blood low shear reduction viscosity and erythrocyte rigidity index, whole blood high shear relative index, whole blood low shear relative index difference between groups (P<0.05); In high dose group of whole blood viscosity, whole blood reductive viscosity, erythrocyte deposited, whole blood high shear whole blood reduction viscosity, whole blood reductive viscosity, relative index difference between groups (P<0.05).Discuss:1. The type of syndrome about chloasma is complicated, but stagnation of liver qi and spleen deficiencycan be summarized as the card type.2. The main reason of chloasma is seven emotions, mainly for modern disorders. The main location of disease are the liver, kidney and spleen. The main nature of disease are deficiency of qi and blood, qi and blood stasis.3. The basic treatment on chloasma can be summarized as tonifying qi and blood, invigorate the circulation of qi. The formulas can be choose with modified xiaoyaosan, liuweidihuangwan, taohongsiwutang, modified shengyutang, and buzhongyiqitang.4. According to the basic principle of formula, syndrome and effect with one accord, MGS is the effect therapy for chloasma; According to the summary about traditional Chinese medical treatment to the chloasma, the blood stasis is the key pathogenesis of chloasma.Conclusion:1. MGS can improve the SOD vitality to suppress the oxidation reaction of chloasma model mice, reduce the formation of peroxides MDA, inhibit tyrosinase’s activity, reduce the distribution of melanin cells in skin, reduce melanin formation and protect cells from tissue damage.2. MGS can reduce blood viscosity, increased blood velocity, a good role for activating blood circulation and improve chloasma state of blood stasis by improving the blood rheology. |
