Cancer was characterered with high disease rate as well as difficult therapy. During the chemotherapy process, anticancer agents were harmful to normal cells and make human body. Therefore, it is urgent to develop a new type of drug with high-efficiency and low-toxicity to cure cancer disease. Some bioactive compounds from plants have the effect to inhibit the growth of bacterial and tumor cells while increase the chemical drug’s activity, so develop a medical plant to cure cancer disease has it’s market.In this paper we studied the extract process of Allium chinense by select solvent from ethanol, methanol, ethyl acetate and water. Consider the extract ratio of Allinase and the toxin, we chose ethanol as our solvent. Then we studied the influence of different time, temperature, concentration and liquid-soild to the extraction of Allium chinense, and take35℃,10%ethanol, extract4h when the liquid-soild ratio was1:1as our extraction condition. The influence of these factors to the extraction of allium chinenese was time>concentration>temperature>liquid-soild ratio.The antitumor bioactivity of the extract against B16cell was also studied. The result showed that the Allium chinense extract has a strong inhibit bioactivity against B16cells, after acting24h, it’s inhibit ratio was57.4%, and IC50was1.78μg/mL. Morphological observation showed that the cell nuclei of tumor cells greatens, growth gap enlarged and the cell shape deformated from spindle to elliptic, the morphological change also showed a concentration and time dependence.The protein of Allium chinense was separated from the extract for further study by Tris-HCl. The protein curve was measured by Brandford.56.4mg Protein was extracted from150g Allium chinense, and the extract ratio was0.0376%. The protein was separated by RP-HPLC and ammonium sulfate. The result showed that the16th peak of the sample separated by HPLC can inhibit the growth of Tritirachium album and the protein separated by80%ammonium sulfate has a remarkable inhibit bioactivity to tumor cells:it’s inhibit ratio against B16and Meth-A cells was75.3%and61.6%at24h,78.5%and66.4%at48h respectively. It’s IC50were11.3μg/mL and13.0μg/mL, respectively。The composition of extract and protein of Allium chinenese was analysised by RP-HPLC and SDS-PAGE, and dected by LC-MS. The result showed that the main compounds of Allium chinense were alliinase, heat shock protein, lectin, cysteine synthase and tumor necrosis factor receptor superfamily member.The composition, molecular structure and faction of Allium chinenese extract and protein needs further research.
