Objective:Recent studies on the association of TNF-a gene-308G>A, IL-10gene-1082A>G polymorphisms with risk of non-Hodgkin’s lymphoma (NHL) showed inconclusive results. Accordingly, we conducted a comprehensive literature search and a meta-analysis to clarify the association.Methods:Data were collected from the following electronic databases:Pubmed, Excerpta Medica Database (Embase), Medline, ISI Web of Science, China Academic Journal database (CNKI), Chinese Biomedical Literature Database (CBM), VIP, Wanfang, with the last report up to Jun,2013. Experiments included in the article are case-control experiments and cohort study, required similar research methods that must consist of the purpose, design and correct statistical methods. Its findings are available to gain odds ratio (OR) and95%confidence interval (95%CI) or other basic data. The quality of the articles included in the study was evaluated by Scale. All the analysis in this study was carried out by R software (Version2.15.1). The OR and its95%CI were used to assess the strength of association. Sensitivity analysis was conducted by repeating meta-analysis with low quality of studies removed. Publication bias were judged by funnel plot and Egger’s test. Results:(1) the association between TNF-a gene-308G>A polymorphisms and the risk of NHL:A significant association between TNF-a gene-308G>A polymorphism and the risk of NHL was observed in the pooled case-control studies (A allele vs. G allele:OR=1.18,95%CI=[1.04,1.34], P=0.0128; GA+AA vs. GG:OR=1.15,95%CI=[1.01,1.31], P=0.0394; AA vs. GG+GA:OR=1.46,95%CI=[1.12,1.90], P=0.005; AA vs. GG:OR=1.51,95%CI=[1.15,1.99], P=0.0031). In the cancer type-based subgroup analysis, we analyzed the DLBCL and FNHL. Similar results were found in the DLBCL subgroup, but there was no evidence of association between TNF-a gene-308G>A polymorphism and the risk of FNHL.(2) the association between IL-10gene-1082A>G polymorphisms and the risk of NHL:A significant association between IL-10gene-1082A>G polymorphism and the risk of NHL was observed in the pooled case-control studies (AG+GG vs. AA:OR=1.16,95%CI=[1.04,1.30], P=0.0073). In the cancer type-based subgroup analysis, we analyzed the DLBCL and FNHL. Similar results were found in the DLBCL (AG+GG vs. AA:OR=1.25,95%CI=[1.01,1.54],P=0.0363; AA+GG vs. AG: OR=0.85,95%CI=[0.73,0.97], P=0.0199)and FNHL (GG vs. AA: OR=3.22,95%CI=[2.40,4.32], P<0.0001) subgroups.Conclusions:(1) TNF-a gene-308G>A polymorphism was associated with susceptibility of NHL, and the allele A might increase the risk of NHL, especially DLBCL.(2) IL-10gene-1082A>G polymorphism maybe associated with the risk of NHL...
