objective: the occurrence and development of colorectal cancer is the resultof many factors. P27is a cell cycle-dependent protein kinase inhibitors (cyclindependent kinase inhibitors, CKI), which regulate the cell cycle to ensurenormal cell division, once the disorder of abnormal cell cycle division willoccur, the tumor will be generated. tumor can not be generated withoutproliferation, when greater proliferation than apoptosis, tumor occurs ordevelop. Ki67can accurately reflect the proliferation of tumor cells, and canhelp determine the benign and malignant tissue. Tumor metastasis andrecurrence is the result of patients with poor prognosis, Tumor recurrence is thetreatment failure for patients, and tumor metastasis can improve early diagnosisand prompt treatment to prevention and control. nm23is now consideredtumor metastasis suppressor gene,whose family has nine members(nm23-H1-nm23-H9),their protein products have nucleotide diphosphate kinase(nucleotide diphosphate kinase, NDPK) activity. NDPK by GDP reduced toGTP,thereby affecting thedepolymerization of microtubules,signal transduction,transcription regulation, and cell adhesion while maintaining the normal stateof the cell division and inhibit metastasis of cancer cells.This research study168cases of colorectal cancer and normal tissues by immunohistochemistryP27, nm23and Ki67expression situation, analyze their relationship withclinicopathological parameters, explore the biological indicators of earlydiagnosis and prognosis, looking for targeted therapy new method.Methods:Collecting84cases colorectal specimens from colorectal cancer surgerypatients, were first diagnosed with primary colorectal cancer from January2013to July2013in Gastrointestinal Surgery, Luzhou Medical College Hospital, allpatients prior to surgery is not for radiotherapy, chemotherapy and other non- surgical treatment. While selecting the appropriate84cases normal tissue fromsurgical resection specimens. All cases have clinical data and pathological data.Immunohistochemistry was used to detect P27Ki67SP and nm23expression incolorectal cancer tissues and normal mucosa, analysising their relationshipbetween clinicopathological factors and with each other.Results:1.P27incolorectal cancer and normal colorectal mucosa specimens positive expressionrates was respectively positive in50%(42/84) and83.75%(71/84), there issignificant difference between the two(P<0.05).low expression of P27incolorectal cancer is connected with the degree of tumor differentiation,lymphnode metastasis and Dukes staging(P<0.05), while cytoplasmic expression inP27was negatively correlated with the degree of differentiation of tumormetastasis, which was also positively correlated with lymph node metastasis.2.Ki67in colorectal cancer, colorectal normal mucosa specimens positiveexpression rates was respectively60.71%(51/84) and10.71%(9/84), there issignificant difference between the two (P <0.05). high expression of Ki67havesomething to do with the degree of differentiation and depth of invasion ofcolorectal cancer(P<0.05), it have nothing to do with age, sex, tumor location,gross type, tumor size, lymph node metastasis, Dukes staging (P>0.05).3.nm23in colorectal cancer, colorectal normal mucosa positiveexpression rates was respectively58.33%(49/84) and83.33%(70/84), there isstatistical significance between the two (P<0.05).low expression of nm23havesomething to do with lymph node metastasis, depth of invasion of colorectalcancer (P <0.05), it have nothing to do with age, sex, tumor location, Dukesstage, tumor gross type, tumor size (P>0.05).There is negative correlationbetween low expression of P27and high expression of Ki67expression incolorectal cancer(r=-0.561,P<0.05),high expression of Ki67and low expressionof nm23is negative correlation (r=-0.383, P <0.05), whereas There is nocorrelation between low expression of P27and nm23in colorectal cancer (P>0.05).Conclusion:1.low expression of P27, nm23and high expression of Ki67 have something to do with the occurrence and development of colorectalcancer.2.The degree of differentiation, lymph node metastasis and Duckesstaging of colorectal cancer is connected with low expression of P27, whilecytoplasmic expression in P27was negatively correlated with the degree ofdifferentiation of tumor metastasis, which was also positively correlated withlymph node metastasis.3. High expression of Ki67have something to do withthe degree of differentiation and depth of invasion of colorectal cancer, lowexpression of nm23have something to do with lymph node metastasis anddepth of invasion of colorectal cancer4.There is negative correlation betweenexpression of P27and expression of Ki67expression in colorectal cancer,expression of Ki67and expression of nm23is negative correlation. detectingP27, Ki67and nm23helps prognosis of colorectal cancer,regulating theexpression of the three cells is a new direction for colorectal cancer targetedtherapy.
